Objective: To establish the portal hypertensive gastropathy (PHG )model through portal vein ligation ,and explore the role of hypoxia inducible factor-1alpha(HIF-1α), vascular endothelial growth factor(VEGF) and pigment epithelium derived factor(PEDF) in portal hypertensive gastropathy through the dynamic expression of HIF-1α,VEGF and PEDF in the gastric mucosa of portal hypertensive rats.Methods : Sixty male Wistar rats were randomly divided into the experimental groups and the control groups. The PHG models were established through one-stage portal vein ligation, and sham-operated rats(SO) were simultaneously established as controls. On the 7th, 14th, 28th day after operation respectively,the portal vein pressure(PVP) was measured,and the expression of HIF-1α,VEGF and PEDF in rat gastric mucosa was detected by immunohistology. Then ,the statistical significance was evaluated by single agent mean square analysis, and the correlation was evaluated by Pearson's correlation.Results: 1)On the 7th, 14th,28th day, the PVP was higher in the PHG groups than that in the SO groups(P <0.01) ,and there was significant differences in the PHG groups(P<0. 05);2)On the 14th,28th day, the expression level of HIF-1αwas higher in the PHG groups than that in the SO groups(P<0.01) ,and there was significant differences in PHG groups(P <0. 05);3) On the 7th, 14th, 28th day, the expression level of VEGF was higher in the PHG groups than that in the SO groups(P<0.01) ,and there was significant differences in the PHG groups(P<0.01);4) On the 7th, 14th, 28th day, the expression level of PEDF was lower in the PHG groups than that in the SO groups(P<0.01) ,and there was significant differences in PHG groups(P<0.01);5) There was positive correlation between the expression of HIF-1αand VEGF (P<0.01), and the Pearson's product-moment correlation coefficient r=0.54.6) There was negative correlation between the expression of VEGF and PEDF(P<0.01),and the Pearson's product-moment correlation coefficient r=-0.55.Conclusion: 1) This model is stable and reliable,it is reliable model-establishment after a period of training and it can be applied in experiment stably;2) The PVP was significantly higher in the PHG groups than that in the SO groups,which indicated that the portal hypertension may affect the expression of HIF-1α,VEGF and PEDF ,and accordingly involved indirectly in the pathogenesis and development of PHG;3) The higher expression level of HIF-1αand VEGF may participate in the pathogenesis of PHG;4) The lower expression level of PEDF may participate in the pathogenesis of PHG;5) The HIF-1αand VEGF may both participate in the pathogenesis of PHG;6) The VEGF and PEDF may both participate in the pathogenesis of PHG...
