Purpose:1. Thromboangiitis obliterans established animal model2. The establishment of animal serum and muscle tissue expression of HIF-1αdetection method3. The study of animal model of thromboangiitis obliterans in serum and muscle tissue expression pattern ofHIF-1α, as thromboangiitis obliterans pathogenesis study provides experimental evidence, which basedtreatment and prognosis of disease detection to provide new ideas.Method:1.Wistar 35 rats were randomly divided into sham operation group, 1 day, 3 days, 1 week, 2 weeks, 3 weeks, 4weeks in the model, n = 5, preoperative serum samples were sacrificing, through to large rat femoral arteryinjection of sodium laurate, manufacture rat model of thromboangiitis obliterans, according to modeling timewere killed, the sham group were killed at 4 weeks, were killed after the former specimens from serumsamples, and take about hind-made mode at the surrounding muscle tissue.2. ELISA assay of serum before and after modeling, the level of HIF-1α, semi-quantitativeimmunohistochemical detection of affected and unaffected sides after modeling muscle tissue expression ofHIF-1α.3. Statistical Methods: The results are used that statistical methods used analysis of variance, pairwisecomparisons using LSD method, all the results obtained by SPSS13.0 software processing.Results:1. Signs of change: the emergence model groups the performance of the left hind limb ischemia, according tomodeling time increases gradually increased 1 day group, 3 days group were left hind limb ischemia, swelling,toe black performance 1 week group show increased ischemia, necrosis extended to the ankle. 2 weeks groupappears dry gangrene, necrotic area to the knee, the local inflammatory reaction, 3 weeks, 4 weeks off theperformance of limb necrosis has serious physical body dry gangrene and even loss. Histopathological said:inflammatory reaction, inflammatory cell aggregation, loss of normal muscle tissue texture, 2,3,4-week groupseen in segmental femoral artery occlusion, extensive intravascular thrombosis, and accompanied by a largenumber of new capillaries. Signs and pathological changes in rats confirmed that rat model thromboangiitisobliterans successfully.2. Serologic test results: All in the model rats were the expression of HIF- 1αin serum were higher than thosebefore, modeling 1 day, 3 days group, 1 weeks, 2 weeks, 3 weeks, 4 weeks, postoperative serum levels of HIF-1αexpression were compared with each group were statistically significant before surgery (P <0.05), before and after the sham operation group, modeling the expression of HIF- 1αin serum was no significant difference(P> 0.05), the serum after the modeling HIF-1αexpression in the rapid increase of 1 day group, 3 days reducedslightly, then gradually decreased, but higher than the level of the sham group, of which between 1 day and 3days, one week and 2 weeks between 3 weeks and 4 weeks was no significant difference between (P> 0.05),remaining in the model were statistically significant comparison (P <0.05).3. Immunohistochemical results: HIF-1αprotein in the sham group and in the model of bilateral lowerexpression in the contralateral muscle, the muscle tissue of the affected side in the model is highly expressed inthe model affected side of the expression of HIF-1αaverage gray value were statistically significant comparedwith the healthy side (P <0.05), of which 1 day, 3 days group, the average gray value of the minimum, nosignificant difference between the two groups (P> 0.05), compared with other manufacturing modules Therewere significant differences (P <0.05), 1 week, 2 weeks expression was no significant difference between thetwo groups (P> 0.05), than the other in the model were significantly different (P <0.05), 3 weeks 4 weeks nosignificant difference between the two groups (P> 0.05), than the other in the model were statisticallysignificant (P <0.05).Conclusion:1. Using the femoral artery injection of sodium laurate produced TAO rat model, the method is simple, a highsuccess rate modeling can be used for scientific research on TAO.2. In this study, developed the initial lesions in the early thromboangiitis obliterans, serum and tissues showedhigh expression of hypoxia inducible factor-1α, as the disease advances are rapidly declining, this may bepatients with TAO and thus failed to establish an effective collateral circulation the direct cause ofexacerbations.
