Oral squamous cell carcinoma(OSCC) is the most common malignant head and neck tumor. It is highly malignant,strong invasion, quickly metastasis and increased incidence result in high mortality rate. Malignant tumor deadly often because itself has strong invasive and transfer ability, which led to multiple organ systemic failure. In recent years, including cell adhesion, protein hydrolysis, migration and angiogenesis interactions, thorough research found the tumor cells invasion and metastasis is a complicated continuous process in which extracellular matrix and basement membrane play a key role for constituting histologic barriers, in limiting the invasion and metastasis of tumor cells. How ever the matrix metalloproteinases (MMP-2) conld degrade collagenⅣwhich is main ingredient of extracellular matrix and basement membrane ,making breakthrough in the primary site of tumor cell and invasion and distant metastasis occurred. It has been found that CD147 exists widely in the human body tissues and organs, also known as extracellular pregnancy-associated matrix metalloproteinases inducing factor, belongs to the immunoglobulin superfamily,studies suggest that as the extracellular matrix metalloproteinase stimulus through many ways stimulate MMPs produce and releas, which could enhancing tumor cell invasion and metastasis ability. Now, the relationship of MMP-2 and CD147 have been the Research focus in the field of Cancer antagonist drugs, which perhaps will provide a new effective index for the treatment of oral squamous cell carcinoma .Objective: This study will examine the expression of CD147, MMP-2 in oral squamous cell carcinoma and normal oral mucosa tissue, combining with clinical pathologic material, discussion t the role and mutual relations in invasion and metastasis.Methods: S-P imrnunohistochemistry was used to evaluate the expression of CD147 and MMP-2 in 38 oral squamous cell carcinoma and 15 normal oral mucosa。 Explore the relationship of expression of MMP-2 and CD147 with differentiation and lymph node metastasis and correlation in the development of oral squamous cell carcinoma.Results:1. Expressions of CD147 and MMP-2 in normal oral mucosa and oral squamous cell carcinomaThe positive rates of CD147 in normal oral mucosa and oral squamous cell carcinoma were 20% and 92.1% respectively, and had the statistical significance(P<0.05). The positive rates of MMP-2 in normal oral mucosa and oral squamous cell carcinoma were 30% and 84.2% respectively, and had the statistical significance as well(P<0.05).2. Relationship between the expressions of CD147, MMP-2 and the pathologic parameters in oral squamous cell carcinomaThere were no statistical significant differences both of the expressions of CD147 and MMP-2 respectively between the groups with different diferentiation degrees in oral squamous cell carcinoma(P>0.05). There was a higher expression level of CD147 in lymph node metastasis group in oral squamous cell carcinoma(P<0.05) compared with that of non-lymph node metastasis group, as well as MMP-2(P<0.05).3. Correlation between CD147 and MMP-2 in oral squamous cell carcinoma In the study, there was a increasing tendency of the expression of MMP-2, which following the gradually increased expression of CD147 in oral squamous cell carcinoma. The spearman rank correlation coefficient was 0.595(P<0.05) and had a positive correlation between CD147 and MMP-2, which supposing that CD147 might be the inducer of MMP-2 to promote its expression.Conclusions:1. The expression level of CD147 is significant higher in oral squamous cell carcinoma than in normal oral mucosa.2. There was no relationship between the expression of CD147 or MMP-2 and the tissue differentiation degree in oral squamous cell carcinoma patients. But both the expression levels of CD147 and MMP-2 had relationship with the incidence of lymph node metastasis closely. CD147 and MMP-2 both played an important role in the invasion and metastasis processes of oral squamous cell carcinoma.3. There was a positive correlation between the expression levels of CD147 and MMP-2 in oral squamous cell carcinoma.
