| This study is to investigate the antimicrobial effect of b lactoferrin (bLf) on Porphyromonas gingivalis(P.g),and the effect of bLf on the activity of trypsin-like protease (TLP) produced by P.g, and try to provide the evidence of the effect of bLf on the treatment of periodontal disease.P.g ATCC 33277 was chosen as the experimental bacterial strain. We used the liquid media two-fold dilution method to study the antimicrobial effect of bLf. The bLf was diluted into ten different concentration,and each mixed with the suspension of P.g (108CFU/ml). After 48h of incubation at 37℃under anaerobic conditions,the minimal inhibitory concentration (MIC) of bLf was determined. Then added different concentrations of bLf into BHI media and also used anaerobic technique to cultivate P.g. Then after determining the absorbance of mixed liquid, the activity of TLP was assayed.The result showed us that bLf inhibited the growth and proliferation of P.g effectively. MIC of bLf was 5mg/ml. The bLf also inhibited the activity of TLP produced by P.g. With the increasing of concentration of bLf, the activity of combined TLP decreased. BLf with high concentration (greater than 1.25mg/ml) could inhibite the activity of free TLP and the low concentration didn't.So wee can got a conclusion that bLf inhibited the growth and proliferation of P.g,also inhibited the activity of TLP of P.g. |
