Dioscoreae rhizoma is a dry root of dioscorea spp.. It has been used as a traditional Chinese medicine and foot for thousands of years. Extracting and purifying the hypoglycemic bioactive components from dioscoreae rhizome to get three important Ingredients:dioscoreae rhizome polysaccharides; dioscoreae rhizome saponins and dioscoreae rhizome flavones. Research and Compare of hypoglycemic bioactive and Antioxidant under model of a-glucosidase, HepG2cells,diabetic mice and achieve the results as follows:1,Extraction and purification of the active position from dioscoreae rhizomeAnalysing of active substance in dioscoreae rhizome absorbed and eluted by macroporous resin can get the results: Although dioscoreae rhizome is treated by above procedures,its activity is not decreased, and this mothed can get rid of Water-soluble polysaccharide effectively. At the same time the initial enrichment of different kinds of Ingredients as further purified source are attained----extraction substance dioscoreae rhizome saponins, dioscoreae rhizome flavones, dioscoreae rhizome polysaccharides.2,research on the glucose-lowering effect made by the dioscoreae rhizome's active site and ingredient.Study on the model constituented by hypoglycemic actiona-glucosidase, HepG2cells and the diabetic mice showing the results:In comparison with that of dioscoreae rhizome polysaccharides, dioscoreae rhizome saponins, dioscoreae rhizome flavones suggested more in inhibitory activity of a-glucosidase in dose-dependent manner. Through the operation of glucose consumption of HepG2cells experiment, compared with dioscoreae rhizome saponins and dioscoreae rhizome polysaccharides, dioscoreae rhizome flavones suggested more in glucose consumption in dose-dependent manner.the result of the diabetic mice experiment demonstrate that Dioscoreae rhizome,Saponins and flavones can decrease the water drinking volume of diabetic mice and level of glucose considerably and help mice gain weight。the reaction of Dioscoreae rhizome is notable. It concludes that three components show synergistic effect among dioscoreae rhizome's dhypoglycemic bioactive based on the experiment.3,Research on the Antioxidation of rhizome's active siteBased on the diabetic mice model to take research of Antioxidation of rhizome's active site and the results shows that Dioscoreae rhizome Saponins could significantly increase SOD activation and decreasing MDA content, dioscoreae rhizome polysaccharides amd flavones have the anti-diarrhoeal activity also. In comparison with one of ingredients, dioscoreae rhizome powder suggested more in inhibitory activity of AR, as well as Antioxidation.
