| Objective:Rheumatoid arthritis is a systemic inflammatory-based multi-autoimmune disease, involving the peripheral joints. Sinomenine clinical application to achieve better treatment of rheumatoid arthritis patients. This research aims to establish a successful model of adjuvant arthritis on the basis of joint by detecting the mast cells of the tissues around joints in rats before and after treatment, serum interleukin-6 changes in order to investigate the treatment of arthritis sinomenine some mechanism.Methods:(1) In the 48 rats, we randomly selected 10 rats as the normal con trol group, named groupâ… . The remaining 38 rats as the experimental group, each rat by intradermal injection of more complete Freund's adj uvant rat model of adjuvant arthritis produced.14 days after modeling, screening out the successful model of 30 rats which were randomly di vided into 3 groups, named groupⅡ~Ⅳ.(2) 14 days after model start,â…¢,â…£groups were orally given TWP tablets sinomenine;â… ,â…¡group were treated with normal saline. (3) The general conditions of rats, me asured at different time points the rats under the foot pad thickness of the ankle joint and the arthritis index score. (4) Cut the ankle immedi ately after blood collection, production paraffin-embedded, hematoxylin eosin staining of a pathological change in its rate, toluidine blue staini ng and degranulation of mast cells in the situation. (5) Measure serum levels of interleukin-6 by enzyme-linked immunosorbent assay. (6) Ma kestatistical analysis by application spss16.0.Results:1 14 days after modeling, in addition to groupâ… , the arthritis index between each group under the foot and ankle showed no significant difference in pad thickness (P>0.05).1 week after administration to 4 weeks (After21~42days),â…¢,â…£group began to decrease in rats under the foot pad thickness of the ankle joint and arthritis index compared with groupâ…¡, the difference was significant (P<0.01);â…¢,â…£were no significant differences (P>0.05).2 The levels of IL-6 ofâ…¡group were significantly higher than groupâ… (P<0.01), after treatment in each group of IL-6 levels lower than that in groupâ…¡(P<0.01),â…¢, IVwere no significant differences (P>0.05).3 Hematoxylin eosin staining showed that compared with groupâ…¡, treatment groups rats with adjuvant arthritis may reduce the pathological changes in joints (P<0.01),â…¢,â…£had no significant differences (P> 0.05).4 Toluidine blue staining showed that compared with groupâ… , the number and degranulation of mast cells of the tissues around joints inâ…¡,â…¢group were both significantly increased (P<0.01),â…¡,â…¢were no significant differences (P>0.05);â…£group mast cell number and degranulation rate of increase has been effectively suppressed, andâ…¡,â…¢group were significantly different (P<0.01).5 Correlation analysis showed that the number and degranulation rate of mast cells in each group, were positively correlated with the joint scores (P <0.01), and the serum levels of IL-6 levels was significantly positively related with the joint pathology scores (P<0.01).Conclusion:1 Sinomenine adjuvant arthritis treatment has a significant effect, can significantly reduce the rat paw edema, improve joint pathological changes.2 Sinomenine treatment of adjuvant arthritis and the possible mechanisms are:(1) regulation of sinomenine have the function of mast cells, by inhibiting the function of mast cells to improve the pathological changes in rat joints, which play on adjuvant arthritis in rats. (2) by reducing cytokines such as interleukin-6 secretion, reducing their joint organization of the stimulus, thereby reducing the symptoms of arthritis and cartilage destruction. |
