1. Objective On the basis of a systematic study on blood serum chemical pharmacy of Chinese herbal compound, the author tries to reveal the correlation between the migration of traditional Chinese medicine in the blood components (including the prototype compound and their metabolites) and the fingerprint pattern of the ingredients of liquid compounds in order to establish evaluation index, the serum fingerprint chromatograms, in the hope that it can better characterize the active ingredient compound and can be used in technical research.2.Methods(1) Research on the 6 effective components from extracts in Shuxiong Tablets:HPLC is adopted; notoginsenoside R1, ginsenoside Rg1 and Rb1 to use acetonitrile-water(containing 0.1% phosphoric acid) as the mobile phase, wavelength 203nm; ligustazine and ferulic acid to use methanol:0.1% phosphoric acid (40:60)) as the mobile phase., wavelength 294 nm; hydroxysafflor yellow A to use acetonitrile:0.1% phosphoric acid(10:90). as the mobile phase, wavelength 403nm.(2) HPLC fingerprints study on the extracts of Shuxiong tablet:HPLC is adopted respectively in the measures on mixed extracts and each single extract, and analysis is made on fingerprint chromatograms.(3) HPLC research on the serum fingerprints chromatograms of Shuxiong tablet extract based on blood serum chemical pharmacy:after SD rats are administrated by gavages, samples are taken respectively after certain periods and measures are made on serum chromatograms of both mixed extracts and each single extract, then analysis is made.3.Results(1) Research on the 6 effective components from extracts in Shuxiong Tablets illustrates that, in the selected chromatographic conditions, the 6 effective components from the mixed extracts of Shuxiong tablet can be well separated from others. The separation was more than Rs 1.5, the average recovery was 98%~102%, RSD were within 2.5%.(2) HPLC research on fingerprint chromatograms illustrates that:among the total 15 fingerprint peaks of the HPLC fingerprint chromatograms, fingerprint peaks of No.1,5,6,11,12,14 are in respective accordance with ligustazine, hydroxysafflor yellow A,ferulic acid,notoginsenoside R1,ginsenoside Rg1 and ginsenoside Rb1; No.11,12,14,15 are in accordance with Panax notoginsen; No.3,4,6,8,13 are in accordance with lung cancer., among which NO.3,4 share a larger scope of the peak, which is difficult to separate with the peak shape as scissors; and peaks No.2,5,7,9,10 are in accordance with Carthamus tinctrius L(3) HPLC research on the fingerprint chromatograms also illustrates that: the best result will be achieved with blood collected 30min after the last administration; 16 components can be detected in the fingerprint serum, including 5 of the serum itself, and the other 11 may come from the original compound and their metabolites, including the 6 major components of Shuxiong tablet (ligustazine,hydroxysafflor yellow A,ferulic acid,notoginsenoside R1,ginsenoside Rg1,ginsenoside Rb1) which are in the blood or transitional elements in the blood. The serum chromatograms of Shuxiong tablet are established on the basis of serum HPLC fingerprint chromatograms and compound HPLC fingerprint chromatograms.4.Conclusion(1) HPLC research on fingerprint chromatograms is reliable, accurate and specific to measure the 6 effective ingredients of the Shuxiong Tablet extracts, and this method can be adopted in dynamic characteristics. (2) HPLC research on fingerprint chromatograms is simple and reproducible, and the fingerprint chromatograms can reflect multi-ingredients in the Shuxiong tablet, therefore, this study provides a reliable method for the effective quality control of Shuxiong tablet, and prepares for in-depth study on the tablet.(3) The serum HPLC fingerprint chromatograms contents much information about the transitional elements in the blood, which can be used for a better quality control. And this method is better in retaining traditional Chinese medicine Compound of the active ingredients and thus enhances the efficacy of traditional Chinese medicine; at the same time, it avoids the inconvenience, high cost and time-consumption of the direct use of fingerprints as the evaluation index.
