With growing concern about the extent of health, Chinese Herbal Medicine Polysaccharides with activities of such immune regulation, anti-infective, anti-tumor and so on, is more and more popular by people.We know that APS, ginseng polysaccharides, LBP, lentinan and total polysaccharides extracted from Sijunzi decoction are common. The role of Chinese Herbal Medicine Polysaccharides are not the same, but most of them play the role of Immune regulation. Lentinan is isolated from the common mushroom extract fruiting bodies, and its mechanism has not clearly reported. This study tries to make a preliminary discussion on the mechanism of T cells by lentinan.1. Experimental Research1.1 Screening of Chinese Herbal Medicine PolysaccharidesIn this study, polysaccharide as the research object, first select some common polysaccharides to be screened, and then focus on the further study of one polysaccharide. Experiments using in vivo and in vitro experiments, with reference to the thymus index, spleen index and lymphocyte transformation stimulation index indicators, and ultimately lentinan was selected for the further study.1.2 Screening of Lentinan and selecting DTH mice modelAfter determing the object for the study of lentinan, its concentration were studide at threre different dose which is from high to low. Found that effects that lentinan of 40mg/ml group were better than others by comperison of indicators. In the choice of animal model, taking into account the effect of lentinan on regulation of the immune system, the focus was on T cell, so we selceted murine DTH model,which is of of T cells associated with. It's convenient for us to make the mechanism of lentinan working.1.3 Effects of T cell ratio and activation in CLN and PPs by lentinanCompearing lentinan group with model group in CLN,T cell ratio and the expression of T cell activation were on the rise. So are they in PPs. It indicates that the chances of T cell ratio and the expression of T cell activation are one way to effect humoral immune by lentinan.1.4 Indicators and methods for detectionChinese Herbal Medicine Polysaccharides were screened by swelling of the ear, spleen and thymus index, the number of PPs and stimulation index of lymphocytes; The best concentration of lentinan was also screened in this way; Flow cytometry was used to detect the ratio of T cell in CLN and PPs,which were marked with CD3 and B220 antibody. So was the exrpession of T cell activation in CLN and PPs, which were marked with CD3CD25 or CD3CD69 antibodies. In the study of serum INF-γlevels, ELISA Kit was uesd,then optical density values measured by microplate reader, and finaly got the content of INF-γ.2 ConclusionEffects of lentinan on Immunocompromised mice in the experiment of DTH were that the content of INF-y was on the rise.which result in the changes on the ratio and the expression of activation of T cell. And it is proved in this study. It indicated that it is possible for lentinan to effect the ratio and the expression of activation of T cell by rising the content of INF-y.The results of the ratio and the expression of activation of T cell are similar in both CLN and PPs suggest that lentinan may enter the body through the intestinal mucosa and then to the body as a whole from the gut. Its mechanism is worth further study.3 Significance of the ResearchIn this study, we had a better understanding of the mechanism of effects of lentian on Immunocompromised mice. It gave more information to other Chinese Herbal Medicine Polysaccharides research and would be better guide to clinical application of Chinese Herbal Medicine Polysaccharides...
