Object:To prepare the total coumarins from dried fruits of cnidium monnieri (TCCM), and to study its quality evaluation and transdermal diffusion characteristics in vitro as well as comparing with two models of absorption that TCCM and Ost. To study the effect of essential oil from cnidium monneri as enhancers on the percutaneous penetration of Ost, and comparing with Azone. The optimum conditions of processing technology were obtained through orthogonal test.Methods:Drug permeation experiments were performed through excised skin with Franze diffusion cells. TCCM concentration in sample was determined by reversed-phase high performance liquid chromatography (HPLC).TCCM containing 0%,1%,3% and 5% azone and0%,3%,5%,8% ESSENTIAL OIL FROM CNIDIUM MONNIERI was respectively self-made. Then 1 g of above TCCM was separately smeared on the ex vivo skin of rabit.The permeability test was performed in a Franz diffusion cell. TCCM concentration in sample was determined by reversed-phase high performance liquid chromatography (HPLC).Result:1 Percutaneous Absorption of TCCMThe flow rate was of TCCM is slightly larger than osthol which use saline-alcohol (7∶3, v∶v) as receive solution. (JTCCM=0.6554μg/cm2*h>Josthol=0.5474μg/cm2*h). Absort equation of TCCM is Qn=0.6554t-0.0281(r=0.9809) and osthol is Qn=0.5474t-0.1337 (r=0.9998).2 Enhanced the transport of TCCM from different concentrations of essencial oil 3%,5%,8%essencial oil of Fructus Cnidii can enhance the cumulative permeation quantity among all time frame (2,4,8,12,24h), the dataes of ER are2.68,2.36,1.77 respectively and enhance the transdermal speed, which arel.7621μg/cm2*h,1.5465μg/cm2*h,1.1568μg/cm2*h respectively, obviously higher than the transdermal speed of TCCM without essencial oil.We can found that The optimum result is 3% essencial oil.3 Enhanced the transport of TCCM from different concentrations of Azone1%,3%,5% Azonei can enhance the cumulative permeation quantity among all time frame(2,4,8,12,24h), the dataes of ER are3.69,3.42,2.70 respectively and enhance the transdermal speed, which are0.8570μg/cm2*h,1.0333μg/cm2*h,1.1146μg/cm2*h respectively, obviously higher than the transdermal speed of TCCM without Azone. We can found that The optimum result is 5% Azone.4 Enhanced the transport of TCCM from different concentrations of Azone and SO3%SO+3%Azone,3%S0+5%Azone,5%S0+3%Azone,5%S0+5%Azone can enhance the cumulative permeation quantity among all time frame (2,4,8,12,24h), the dataes of ER are2.57,3.10,3.35 respectively and the transdermal speed is obviously higher than the transdermal speed of TCCM without Azone and S0.We can found that The optimum result is 3%S0+3%Azone.Conclusion:permeability and absorbability of TCCM and Ost have no significant differences. no synergistic effect has shown in percutaneous absorption of TCCM. It is very necessary for us to further make an inquiry. The permeation enhancing effect of fructus cnidii volatile oil and Azone has equal absorption intensity of metronidazole, and when used in combination, the enhancing effect is strong and eviden.
