Objective:This study was designed to investigate the effects of clostridium difficile toxin A on anti-proliferation of K562 cells and its mechanism.Methods:The proliferative activity of K562 cells exposed to Ted A was tested by MTT assay. Cell cycle distribution and mitochondrial membrane potential were analyzed by flow cytometry. The protein expression of cytochrome C was observed by immunohistochemistry staining. DNA fragmentation was observed by agarose gel electrophoresis.Results:Toxin A inhibited proliferation of K562 cells in a time and concentration dependent manner. Cell cycle was prevented in Go/G1 phase. Peak of early apoptosis was discovered. The protein expression of cytochrome C increased compared with control group (p<0.05). Agarose gel electrophoresis of DNA from K562 treated with toxin A revealed a "ladder" pattern.Conclusion:Clostridium difficile toxin A could inhibit proliferation and induce apoptosis of K562 cells. The mechanism may be that mitochondrial membrane potential broke down. Mitochondrial matrix released cytochrome C that can induce apoptosis.
