The Influence Of Alpha Defense To Herpes Simplex Keratitis

Objective:Through the establishment of herpes simplex keratitis in mice, observe distribution and change of alpha defense in mice model , and establish people corneal epithelial cells of herpes simplex virus infected cell model, to observe the therapeutic effect of alpha defense to herpes simplex keratitis , research on the influence of alpha defense to herpes simplex keratitis.Methods :Herpes simplex keratitis mice model of Balb/c: select 90 of the inbred line Balb/c mice, randomly divided into three groups according to random number : normal controls group : 10, don't do anything.control group: 40,only do"十"scratch to this group,don't vaccinated herpes simplex virus.Infection herpes simplex virus groups:do"十"scratch on those rats and vaccinate herpes simplex virus,knead 30s.View binformation of rats after the operation by slit-lamp biomicroscope;Do HE pathological observation about inflammatory condition after 3,5,7,15,30day.Immunofluorescence to detect the disposition of HSV-1 andα-defense in corner after 3,5,7,15,30day.The model of cell:chose the human cornerepithelial cell vaccinate in 96 orifice plate,each orifice 100μl,put into incubator of 37℃,5%CO2 until covered the floor.Add HSV-1 andα-defense into the orifice at different time,observation the Cytopathamic effect(CPE).Result:1,Mice were observed by slit lamp,after 7 days they were Significant morbidity,Eyelid swelling, hair removal, corneal epithelial sodium fluorescein staining colored linear or dotted;After 15 days,cornea recovered transparent ,Sodium fluorescein staining and color;After 30 days ,See section scar mouse corneal stroma.Corneal"十"scratches method to establish the herpes simplex keratitis mouse model can be used for research of alpha defense , have good maneuverability, repeatability.2,postoperative mice by slit-lamp biomicroscope , postoperative 7 days, invasion obviously,eyelid edema,hair removal,dyg by fluorescite we can see coloration like dot and line. postoperative 15 days, corneal recovery clearing almost and have no coloration, postoperative 30 days part of the stromal layer of cornea have scar .3,Immunofluorescence :After infection with the changes of corneal are different degree of fluorescence,the fluorescence is significant postoperative 7 days and fluorescence can be see at corneal epithelium,cornea propri,corneal endothelium.The disposition of fluorescence in corne is the same betweenα-defense and HSV-1.4,Observation the role of alpha defense at corneal epithelium cells about herpes simplex virus infection by CPE.Findα-defense in the direct inactivated HSV - 1 and biosynthesis aspects which after HSV - 1 infection have more apparent effect.But it have no work about protection of HCEC befor infection.With the extension of observation time,the trend of result about CPE become to the same compare with the group of infect HSV-1 only.Conclusion:1,Corneal"十"scratches method to establish the simplex keratitis mouse model can be used for research of alpha defense , have good maneuverability, repeatability.2,α-defense have no expression at normal corea .But after infection,the expression and disposition have deep relationship with HSV-1.3,α-defense can inactivate HSV - 1 derectly and inhibit biosynthesize about HSV-1.Butα-defense can not degrade the susceptivity about HSV-1.In other words ,it can not obviate HSK.
