| Object:To establish the method of the model of slow transit constipation (STC)by irrigating the rat's stomach with Diphenoxylate and investigate significance of PGP9.5,SS,AQP3 and VIP expression in proximal colon of the STC in rats. To explore the mechanism of Biansetong Mixture in the treatment of constipation.Methods:70 health SD rats were used in the study. They were assigned randomly into control group,model group, low dose Biansetong Mixture, middle dose Biansetong Mixture, high dose Biansetong Mixture, MaRen Pill and mosapride groups with 10 rats in each group. Control group was irrigated with 0.9%Nacl. Model group and treatment group were used to choose the fittest dosage of Diphenoxylate(10mg/kg) by irrigating the rat's stomach to establish the slow transit constipation model. After the establishment of the model, the treatment group were respectively given low dose Biansetong Mixture, middle dose Biansetong Mixture, high dose Biansetong Mixture, MaRen Pill and mosapride. To observe stool grains,stool weight(wet weight) and the changes of histology. To compare PGP9.5,SS,AQP3 and VIP expression by the immunohistochemistry staining in proximal colon. The staining results were analyzed semi-quantitively by computerized color image analyzer using the following parameters: mean density (MD).Results:(1) The stool grains of the model group was significantly less than that of the control group(11.8±2.2 vs 22.3±1.9, P<0.05),and there was a significantly decrease in stool weight daily(2.19±0.31 vs 4.20±0.14, P<0.05) . BST mixture improved stool grains,stool weight(wet weight) with slow transit constipation. BST is superior to Maren Pill and mosapride in all the above mentioned aspects(.2)The colon tissues in control group is normal. In model group ,inflammatory cell infiltration were observed in colon tissues. Vacuolar degeneration was observed in myenteric nerve plexus and neural cells decreased. BST mixture decreased inflammatory cell infiltration and vacuolar degeneration in colon tissues. (3)The MD values of PGP9.5 was significantly lower in the model control group than the control group (0.116士0.039 vs 0.423士0.024, P < 0.05). The MD values of SS, AQP3 and VIP were higher in the model control group than the control group(0.331士0.034 vs 0.098士0.033;0.370士0.045 vs 0.115士0.029;0.223士0.025 vs 0.077士0.044,P < 0.05). BST mixture could obviously depress the expression of SS,AQP3,VIP ,but it could up-regulate expression of PGP9.5 in proximal colon (P<0.05).(4)The effect of the high-dose and middle-dose groups were superior to the low-dose group of BST group. These effects were highly dependent on a certain dose-effect relationship. Drug treatment group was significantly better than the model group.Conclusion:BST mixture is invigorating qi to strengthen spleen,ascending lucidity and descending turbidity,harmonizing spleen and stomach. It has an obvious facilitation function to the STC rats colon, which can help the STC rats defecating. The colonic emptying promoting effects of BST may be correlated to the decreased contents of SS,AQP3,VIP and the repaired of neural cells in colon tissues. |
