| Objective The main changes in knee osteoarthritis is the degeneration of articular cartilage and the bone hyperplasia of joint margin, on which there is no effective clinical measures, and it has a serious impact on the lives of patients. Early treatment of knee osteoarthritis is of great practical significance. AQP3 plays an important role in the pathogenesis of osteoarthritis. In this study, rabbit osteoarthritis model was replicated by fixing their knees straightenedly in plaster cast, to observe the effect of low-frequency pulsed ultrasound on the expression of articular cartilage and synovial aquaporin3 and the protective mechanism on articular cartilage and synovial in rabbits knee osteoarthritis from the form, function and other aspects, and to provide preliminary experimental evidence for looking for a safe, effective method of treatment for the clinical.Methods Experimental osteoarthritis was replicated in knees of thirty-six mature New Zealand rabbits by fixing their knees straightenedly in plaster cast. They were divided into two groups randomly, eighteen rabbits each group. The experimental animals received IFPUS therapy, of which the intensity was 2.0W.The IFPUS therapy was administered once a day and lasted for 15 min every time. For the control group, sham US therapy was administered (the procedure was the same as the experimental group, but the ultrasonic machine was off and no energy was exported from the detecting head). The experimental animals were randomly sacrificed at 2 weeks,4 weeks and 8 weeks after therapy and 12 rabbits were sacrificed for each time. The immunohistochemistry technology was applied to detect the expression of articular cartilage and Synovial AQP3 after therapy and the results were analyzed statistically.Results With the time prolonged after modeling, the expression of articular cartilage and synovial AQP3 in each group were increased, and the expression of articular cartilage and Synovial AQP3 in the treatment group was lower than that in the control group, and all the differences between the two groups were statistically significant(p<0.01).Conclusion:1. The rabbit knee osteoarthritis model is established successfully.2. Aquaporin 3 may be involved in pathophysiologic process of osteoarthritis.3. It is suggested that the low-frequency pulsed ultrasound might reduce the expression of AQP3 in articular cartilage and synovial at protein leve1, relieve joint swelling, reduce the severity of articular cartilage and synovium degeneration, and have a protective effect on cartilage and synovial. |
