Objective:To investigate the multidrug resistant reversing effect and its possible mechanism of curcumin on human colon cancer cell line HCT8/VCR.Methods:By MTT colorimetric assay,the dose-effect relationship of curcumin on colon cancer cell line HCT-8/VCR was determined to select the appropriate dose of curcumin, the growth curve of cells was draw to evaluate the effect of curcumin on cell growth and proliferation and the drug sensitivity analysis was conducted to observe the effect of curcumin on cell drug sensitivity; Rhodamine123 efflux was performed to detect P-gp transpoter activity of cells treated with curcumin; RT-PCR assay was applied to determine the changes of the mdr1 mRNA of multidrug-resistance gene and Western blot assay was applied to test the changes of the expression of the mdr1 gene products P-gp.Results:The study have shown that curcumin without obvious toxicity to colon cancer cell line HCT-8/VCR under the dose of 25μmol/L, but the growth of cells was sigmificantly inhibited. its toxicity showed dose effect once over this concentration; the sensitivity of cells to VCR,DDP,5-FU,HCPT was enhanced, intracellular Rh123 accumulation was increased (P<0.05), and the expression level of the mdr1 mRNA and protein was significantly suppressed(P<0.05).Conclusion : The study suggest that curcumin can inhibit the expression and function of drug resistance genes, and increase the sensitivity of colon cancer cell line HCT-8/VCR to chemotherapeutic drugs, so as to reverse the multidrug resistance of colon cancer cells, it is worth to do more in-depth and systematic pre-clinical studies.
