Object:To establish chronic cerebral ischemia rat model and to explore the mechanisms of shenlongjiannao capsule which can inhance the learning and memory ability of chronic cerebral ischemia rats.Method:The chronic cerebral ischemia model rats were established by two-vessel occlusion.50 rats were randomly divided into 5 groups:sham operatio-n group,model group, shenlongjiannao capsule groups(high and low dose) a-nd positive control group of pisacetam.After treated by shenlongjiannao ca-psule and pisacetam for four weeks,the learning and memory function was tested with Morris water maze.The contents of nitric oxide(NO),inducible ni tric-oxide synthase(iNOS),hydroxyl free radical(·OH),superoxide dismutase(SO D),malondialdehyde (MDA) and acetylcholinesterase(AchE) in cerebral cort-ex were measured,then the total proteins in hippocampus were analyzed by two-dimensional gel electrophoresis(2-DE).The 2-DE drafts were analyzed b-y ultraflexTOF/TOF mass spectrometry.In the end,the different protein infor-mation were got through NCBInr database.Result:1 The chronic cerebral ischemia rat model was successfully established by two-vessel occlusion.The data of Morris water maze experiment displayed that compared with sham operation group,the escape latency of model group were significantly prolonged(P<0.01),while in the shenlongjiannao capsule group(high and low dose),the escape latency were obviously shorter than that of model group. (P<0.05, P<0.01)2 Shenlongjiannao capsule can reduce the contents of NO,iNOS, MDA,·OH and increase the activity of SOD,AchE.There were significant differences between shenlongjiannao capsule groups and model group(P<0.05 P<0.01).3 The differences protein expression in hippocampus between model group and shenlongjiannao capsule (high dose) were analyzed by proteomics mea-ns.Compared to model group,the expression of 2 proteins were up-regulated and that of 6 proteins down-regulated in shenlongjiannao capsule group(high dose).As a result,8 proteins were identified by ultraflex TOF/TOF.These dif-ferent proteins include tubulin proteins,enegy-related proteins and heat-shock protein,etc.Conclusion:Shenlongjiannao capsule can inhance the learning and memory ability of cerebral ischemia rats.The mechanisms of shenlongjiannao capsule may be:abating the toxicity of NO,inhibiting oxidative stress,improving the function of cholinergic system in cortex and affecting the expression of some proteins in hippocampus.
