| Objections:Varicose veins is a common surgical disease, its treatment are surgery, laser, radio frequency, foam sclerotherapy, including foam sclerotherapy for its efficient, fast, safe, inexpensive features a phlebology in the field of the last 10 years the most important development one. Many different types of sclerosing agent on the market, Ju Gui Chun is the more commonly used domestic sclerosing agent. To assess the efficacy and side effects of Ju Gui Chun, this study utilize the animal vein after different selerosing agent injeetion,through the immunohistochemieal evaluatation of VEGF and conventional HE staining of vaseular ,to observe the impact of blood vessels after different sclerotherapy,so as to the choice of relatively ideal seleroing agent,to provide a theoretical basis.Materials and methods:Experimental animals used 52 New Zealand white rabbits, male and female no limited, weight 2.32±0.23kg, were randomly divided into 4 groups, Ju Gui Chun injection group(n=13),5% sodium morrhuate group(n=13),absolute alcohol injection group(n=13) and physiological saline group(n=13). According to the length of injection time points,13 groups 2h,4h,6h,8h,15h,24h, 36h,2d,3d,4d,5d,6d, 1w. Animals were sacrificed in accordance with the following time:2 hours,4 hours,6 hours,8 hours 15 hours 24 hours 36 hours 2 days 3 days 4 days 5 days 6 days,7 days after injection, the blood vessels do conventional HE staining and immunohistochemical staining to evaluate the efficacy and side effects of the curing agent, and tissue repair time.Results:After three vein sclerotherapy can be found in HE staining of vascular endothelial cell injury, the most important of which absolute alcohol, Ju Gui Chun and 5% sodium morrhuate is similar to the second. Determination of the minimum time for experiments within 2 hours of group Ju Gui Chun and 5% sodium morrhuate groups occur:aseptic inflammation, vein endothelial cells (VEC) destruction,proliferation, alignment is not continuous, part of the vascular endothelial cell loss, swelling, vascular endothelial cell nuclei Stained and larger. Venous VEGF immunohistochemical staining using integral evaluation, Ju Gui Chun group and 5% sodium morrhuate group showed increasing trend, respectively, at the 4 th day and reached a peak 3 th days. In the absolute alcohol group showed a downward trend, becoming the 6th day 0. Ju Gui Chun injection group, Ju Gui Chun injection group and absolute alcohol injection group sessions in each group comparison p values were less than 0.01, there was significant difference. physiological saline injection group compared in each group no significant difference in time. Ju Gui Chun injection group, Ju Gui Chun injection group, absolute alcohol group and the physiological saline injection group and any two of each group contrast p value less than 0.01, significant statistical difference.Conclutions:According to pathological changes seen from HE staining,as well as the expression of VEGF in animal tissue specimens after different scleroing agent injection,HE staining showed Ju Gui Chun have some characteristics, such as Intimal aseptic inflammation and blood vessels injury small, thrombosis fast and organizations reeovery quick,VEGF espression in 4rd day reaeh summit, histology and the expression of VEGF consistent. Confirmed that it is a more ideal sclerosing agent. HE staining showed Absolute alcohol have some feature that thrombosis fast,tissue injury large and tissur repair slow,VEGF expression and HE staining were consistent,6 th -7th day almost no expression. Suggesting that its should not be used as clincical application of conventional sclorsing agent to treatment varicose veins. HE staining showeds 5% sodium morrhuate have good embolization results but the blood vessels injury more serious than Ju Gui Chun,VEGF expression showed the process of mild rise then significantly decreased,tissue repair between Ju Gui Chun and absolute alcohol, there is still space for clinieal application. |
