| Objective: Selected BushenZhuyun Recipe and Xiaoyao Pill as representative prescription of Bushen and Shugan treatment. Based on the experimental evidence of improving the endometrial receptibility function with Bushen and Shugan treatment, we study the comparative effect of BushenZhuyun Recipe and Xiaoyao Pill in endometrial LPAR-3 and Galectin-3 of endometrial receptibility obstacles rat models. We try to investigate the mechanism and role of Bushen and Shugan treatment in improving the endometrial receptibility, enrich Traditional Chinese Medical theory about"the liver is the innate of women"and"Kidney Plays an important role in the reproductive", and promote the development of Chinese medicine of gynecology.Methods: Chosen 140 healthy female KM mice (weighing 25-30g) and 30 healthy male KM mice (weighing 39-35g). Based on the methods of Pan Yong-miao, Zhang Han-wang Inc, having Vaginal epithelial cells smear to observe estrous cucle once a day for two weeks. 102 mice with regular estrous cucle were randomly divided into six groups: one normal group, one model group and four treatment groups (BushenZhuyun Recipe high-dose group, BushenZhuyun Recipe low-dose group, Xiaoyao Pill high-dose group, Xiaoyao Pill low-dose group), the treatment groups were drenched with BushenZhuyun Recipe or Xiaoyao Pill in high-dose (5.4 g/ml/is 100 gold weight, equivalent to clinical doses of 24 times) or low-dose (2.7 g/ml/is 100 gold weight, equivalent to clinical doses of 12 times , weighed mice every week, adjusted the delivery quantity, each group was made up of 17 mice. The treatment groups and the model group were given intraperitoneal injection of Alarelin (40IU /100g) In post estrus once a day for 9 days and the ninth day received an injection of HMG (1IU /100g), 48 hours after the injection, injected HCG (10IU /100g) peritoneal. The normal group only received an injection of same amount of saline. Irrigated herbs (Bushen Zhuyun Recipe or Xiaoyao Pill) to the four treatment groups once a day for 11 days. The normal group and the model group received a Gastric lavage of same amount of aqua destillata once a day.The female KM mice were mated with the male KM mice at the rate of 2:1 at the night that the model group and the treatment group were given HCG intraperitoneal injection and the normal group was the ovulation. The next day, searched the female mice'vaginalplu, chosen the pregnant mice to do the experiment. After the fourth day'midnight of the mice'pregnancy, Cut head to execute mice, took the left uterus by Aseptic caesarean, Add EP tube, liquid nitrogen, then quickly turn into - 80℃cross-matched, fixed them in EP tube, and then Put them into liquefied nitrogen for long term Preservation to do RT-PCR, observed LPAR-3 and Galectin-3; fixed the right uterus in 4% Para formaldehyde solution to do Immunohistochemistry, observed LPAR-3 and Galectin-3.Results:1 The effect of BushenZhuyun Recipe and Xiaoyao Pill to the expression of the endometrial LPAR-3 Protein and mRNA in endometrial receptibility obstacles rat models.Compared with normal group, LPAR-3 Protein in model group was signify-cantly decreased(P<0.01). Compared with model group, LPAR-3 Protein in BushenZhuyun Recipe low-dose group,Xiaoyao Pill low-dose group were increased(P<0.05). (Table 1)Compared with normal group, LPAR-3 mRNA in model group was decreased(P < 0.05). Compared with model group, LPAR-3 mRNA in BushenZhuyun Recipe high-dose group,Xiaoyao Pill high-dose group,low-dose group were increased(P<0.05),Xiaoyao Pill high-dose group were returned to normal group(P>0.05).(Table 2)2 The effect BushenZhuyun Recipe and Xiaoyao Pill to the expression of Galectin-3 Protein and mRNA in endometrial receptibility obstacles rat modelsCompared with normal group, Galectin-3 Protein in model group was significantly decreased(P<0.01). Compared with model group, Galectin-3 Protein in BushenZhuyun Recipe high-dose group,low-dose group were significantly increased(P<0.01).BushenZhuyun Recipe high-dose group was returned to normal group(P>0.05).BushenZhuyun Recipe high-dose group and low-dose group had no significant difference(P>0.05). Compared with model group, Galectin-3 Protein in Xiaoyao Pill high-dose group,low-dose group were significantly increased(P<0.01).Xiaoyao Pill low-dose group was returned to normal group (P>0.05).Xiaoyao Pill high-dose group and low-dose group had no significant difference(P>0.05).(Table 1)Compared with normal group, Galectin-3 mRNA in model group was significantly decreased(P<0.01).Compared with model group, Galectin-3 mRNA in BushenZhuyun Recipe high-dose group was significantly increased(P < 0.01).Xiaoyao Pill high-dose group was increased(P <0.05)(Table 2).3 The differences effect between BushenZhuyun Recipe and Xiaoyao Pill on Protein and mRNA in endometrial receptibility obstacles rat modelsCompared with BushenZhuyun Recipe low-dose group, Galectin-3 Protein in Xiaoyao Pill low-dose group was significantly increased(P<0.01).Galectin-3 Protein in BushenZhuyun Recipe high-dose group and Xiaoyao Pill high-dose group was no significant difference(P>0.05).LPAR-3 Protein in BushenZhuyun Recipe group and Xiaoyao Pill group was no significant difference(P>0.05).(Table 3)mRNA in BushenZhuyun Recipe high-dose group and Xiaoyao Pill high-dose group was no significant difference(P > 0.05).mRNA in BushenZhuyun Recipe low-dose group and Xiaoyao Pill low-dose group was no significant difference(P>0.05).(Table 4)Conclusions:1 BushenZhuyun Recipe and Xiaoyao Pill can both improve LPAR-3 and Galectin-3 in endometrial receptibility obstacles rat models.2 Galectin-3 Protein in Xiaoyao Pill low-dose group was superior to BushenZhuyun Recipe low-dose group, Galectin-3 Protein in BushenZhuyun Recipe high-dose group and Xiaoyao Pill high-dose group had no deference.3 LPAR-3mRNA in BushenZhuyun Recipe high or low dose groups had no deference with Xiaoyao Pill high or low dose groups. |
