| BackgroundProstate cancer(PCa) is one of tumor that serious threat to the health of the old age male,morbidity of Prostate cancer increased noticeably in global scope, The incidence rate of PCa is the third of all kinds of male tumors,The fatality rate is sixth.Invasion and metastasis of tumor cells are the primary causes of fatal outcome of cancer diseases.To control tumor cell's invasion and metastasis is still an important topic in today's tumor research.Reasearch manifest that intercellular adhesion might be an important factor in generation,development,invasion and metastasis of cancer, E-cadherin (E-cad)is one of the most important cell adhesion molecules, The study about intercellular adhesion in the molecule mechanism of tumor metastasis is an important medical scientific reseach.Internal and overseas,the role of E-cad in the Prostate cancer's generation,development,invasion and metastasis is not yet identify report. ObjectiveThis study examined the expression of E-cadherin in Prostate cancer tissue and study the serum levels of soluble E-cadherin in patients with Prostate cancer,to analysis the relationship between expression E-cadherin,serum levels of sE-cadherin and clinicopathologic features, explore the values of diagnosis, invasion,metastasis,prognosis in Prostate cancer, analysis the correlation of E-cadherin and sE-cadherin,in order to provide theories based on Prostate cancer. Methods113 patients were collected for disease of prostate with difficulty of urination, Affiliated Hospital, Affiliated KaiLuan Hospital, North China Coal Medical College during January 2007 to December 2008, 42 cases of Prostate cancer,71 cases of Benign prostate hyperplasia,which were confirmed by pathology. All cases were selected by strictly screening. Preoperative ,Venous blood samples(5ml) were collected into plain tubes, and allowed to clot, room temperature standing 1 hour, they were centrifuged at×800g for 10 minutes at 4℃, The serum was removed, aliquoted, and stored at -70℃until it was assayed.Serum specimen of Health control group were collected same as above.Tissue specimens were fixed by 10% formalin liquid, paraffin-embedded to wait for using.Expressions of E-cadherin in these specimens were detected by the Immunohistochemical dicho-step method. The concentration of soluble E-cadherin was measured with a commercially available sandwich enzymelinked immunoadsorbent assay.To collect numerus of TPSA about Prostate cancer. Finally, statistics analysis were carried out.Results1.Positive rate of E-cadherin in Prostate cancer group was 45.24%,which is lower than Benign prostate hyperplasia(94.37%),the difference was significant(P=0.000);There had no significantly correlation between the E-cadherin positive expression and clinicopathologic classification(P>0.05); The low expression of E-cadherin was correlate clinical stage(P=0.020).2.The serum concentrations of soluble E-cadherin in Prostate cancer group(12.4860±6.1731μg/L)were significantly higher as compared to the Benign prostate hyperpiasia group (6.2275±1.9518μg/L),The difference was significant (P<0.05); There had no significantly correlation between the serum concentrations of soluble E-cadherin and clinicopathologic classification(P>0.05); The serum concentrations of soluble E-cadherin was correlate clinical stage(P<0.001).3.The sensibility, specificity, positive prediction value, negative prediction value and diagnose accordance rate in diagnosis of Prostate cancer were 80.95%,78.87%,69.39%,87.50%,79.65%. 4.Positive rate of the expression of E-cadherin was negatively correlated with the serum level of soluble E-cadherin(P<0.05). ConclusionsThe down regulation about expression of E-cadherin may play an important role in the generation,development,invasion and metastasis of Prostate cancer,which may be markers of poor prognosis;The soluble E-cadherin might be a potentially valuable tumor markers to monitoring of the Prostate cancer; The detection about expression of E-cadherin and serum concentrations of soluble E-cadherin can help to evaluate the biological behavior and invasion,metastasis,prognosis of Prostate cancer. |
