| Objective:To study the respective of P120 catenin( P120ctn) and Epithelium-cadherin(E-cadherin) in normal cervix epithelium(NCE), cervical precancerous lesions(cervical intraepithelial neoplasia grade II,III(CIN II,III)), Squmous carcinoma of the cervix(SCC), and to analyz the correlation between the two Protein, and to explore the mechanisms of the both Protein in cervical squamous cell carcinoma.Methods:Clinical data were collected, and the cervical tissues were divided into the normal cervical tissue group(13 cases), CIN group(16 cases, including 8 cases of CIN II, CIN III 8 cases), and SCC group(45 cases, including 15 cases of high differentiated squamous cell carcinoma tissue, 15 cases of medium differentiated squamous cell carcinoma tissue, 15 cases of low differentiated squamous cell carcinoma tissue). According to the clinical data of characteristics(such as age) and clinical pathological features(FIGO stage, depth of invasion, tumor size), SCC group was divided into different groups. Immunohistochemical(IHC) Envision was used to measure the expression of P120 ctn and E-cadherin in different groups respectively. The correlation were analysed between the expression of P120 ctn and E-cadherin in cervical carcinoma and the clinicopathological parameters, and a preliminary study were made on the mechanism of action and relationship in the invasion and metastasis of cervical squamous cell carcinoma. Chi-square test and Fisher ’s exact probability method were used to calculate the experimental results. Spearman rank correlation analysis was used to analyse the correlation. P<0.05 was statistically significant.Results:1. In normal cervix and CIN, P120 ctn and E-cadherin showed strong positive expression of epithelial cell membrane. In cervical squamous cell carcinoma, P120 ctn and E-cadherin were decrease or absent expression in cell membranes,and P120 ctn showed abnormal expression in the cytoplasm.2. The positive expression rate of P120 ctn in normal cervix group, CIN group and SCC group were 100%, 100%, 62.22%. P120 ctn showed expression in normal cervix and CIN tissues, but the expression in cervical squamous cell carcinoma showed a downward trend, and the difference was statistically significant among the three groups(χ2=25.274, P<0.001). There is no significant difference between normal cervix group and CIN group,χ2=5.759,P=0.052; Statistically significant difference between CIN group and SCC group, χ2=20.236,P<0.001. The normal cervix group compared with SCC group, with significant difference, χ2=8.639,P=0.027. Patient age, tumor size, FIGO stage, parametrial invasion and differentiation degree in Cervical squamous cell carcinoma were no correlation with the expression of P120ctn(P>0.05).But the depth of stromal invasion, vascular invasion and lymph node metastasis in Cervical squamous cell carcinoma were correlation with the expression of P120ctn(P<0.05).3. The positive expression rate of E-cadherin in normal cervix group, CIN group and SCC group were 100%, 100%, 75.56%. E-cadherin showed expression in normal cervix and CIN tissues, but the expression in cervical squamous cell carcinoma showed a downward trend, and the difference was statistically significant among the three groups(χ2=23.075,P<0.001). There was significant difference between normal cervix group and CIN group, χ2=10.631,P=0.002. Statistically significant difference between CIN group and SCC group, χ2=9.018,P=0.018; The normal cervix group compared with SCC group, with significant difference, χ2=13.157,P=0.002. Patient age, tumor size,and FIGO stage in Cervical squamous cell carcinoma were no correlation with the expression of E-cadherin(P>0.05).But the depth of stromal invasion, parametrial invasion,differentiation degree,vascular invasion and lymph node metastasis in Cervical squamous cell carcinoma were correlation with the expression of E-cadherin(P<0.05).4. There was positive correlation between the expression of P120 ctn and E-cadherin in squamous carcinoma of the cervix(rs=0.303,P=0.043).Conclusion:1. The expression of P120 ctn and E-cadherin in normal cervix, CIN and SCC decreased, indicating the two protein may be related to the occurrence and development of cervical squamous cell carcinoma related.2. In SCC group, the low expression of P120 ctn was related with the depth of stromal invasion, vascular invasion and lymph node metastasis, suggesting that activation of P120 ctn gene may be involved in the occurrence, development of cervical squamous cell carcinoma, illustrating that P120 ctn play an important role in the invasion and metastasis of cervical squamous cell carcinoma.3. In SCC group, The low expression of E-cadherin was related with the depth of stromal invasion, parametrial invasion,differentiation degree,vascular invasion and lymph node metastasis. The results suggest that E-cadherin also played an important role in the occurrence, development, infiltration and metastasis of cervical squamous cell carcinoma.4.There was Positive correlation between the expression of P120 ctn and E-cadherin in squamous carcinoma of the cervix. The result suggested that the two proteins in the occurrence and development of cervical squamous cell carcinoma may have a common mechanism of action or synergistic effect. |
