The extraction and separation of active ingredients from Chinese herbal medicine play a vital role on the road of modernization of traditional Chinese medicine. In this paper, the methods of extracting of active ingredients from Huperzia serrata and radix linderae are studied. And the separation of Huperzine A is investigated.Analysis of Huperzine A. by HPLC. Chromatographic conditions: mobile phase: methanol: 0.08mol/L ammonium acetate buffer (40: 60); detection wavelength: 308nm; injection volume: 10μl; velocity of flow: 1ml/min; column temperature: room temperature. The standard curve for concentration of Huperzine A vs absorbance was established. By linear regression, the linear range is 0.5μg/ml~16.0μg/ml. The linear equation is: which correlation coefficient is 0.99911. Y=21303.70931 X?12102.70945By comparison of various extraction methods, Microwave-assisted extraction is selected. Basis to single-factor experiment, optimizing the process parameters (concentration of hydrochloric acid, extraction time, ratio of liquid to material) to increase recovery and purity of Huperzine A by RSM. The optimized process parameters are: concentration of hydrochloric acid: 2.03%, extraction time: 20.04h, ratio of liquid to material: 19.22. The theoretical recovery and purity are 85.67% and 3.04%. In order to verify the accuracy of the model, validation test is implemented, and the result are: recovery: 85.31%, purity: 3.05%.Separating of Huperzia serrata extract by silica gel chromatography. And optimized parameters are: mobile phase: methanol: cyclohexane (4: 96); injection volume: 8mg/ml silicone; velocity of flow: 3ml/min. In this condition, the recovery and purity are 77.65% and 48.13%.By comparison of steam distillation and solvent extraction, SFE is selected. Optimizing the conditions by orthogonal test, the results show that: extraction pressure is the key parameter and others are minor. The optimized parameters are: extraction pressure: 30MPa; extraction temperature: 40℃; separation pressure: 4MPa; separation temperature: 50℃. In these conditions, the oil yield is 4.88ml/100g Raw material.
