| Huperzia serrata is traditional Chinese medicine which has been used for thousands of years as a diuretic, hemostyptic, antispasmodic, and analgesic agent. In recent, huperzine A, one of the chemicals in huperzia serrata, has been studied extensive years, for its potential in treating dementias such as Alzheimer's disease.Synthesis of huperzine A and its numerous analogues has been reported. Due to the high cost, however, extraction from plants is still the major source of huperzine A. In this paper, based on the conventional methods of extracting and isolating natural products from Chinese herbs, the improved and optimized process for the purification of HupA from huperzia serrata was established.This paper started with develepment of a rapid, precise and reproducible routine HPLC assay for keeping track and quantification of the compounds through the separation process, And applied to the determination of huperzineA in different parts of Huperzia. Using Symmetry C18 column with Acetonitrile:0.028M KH2PO4(12:88V/V) as mobile phase. The calibration curve was linear in the range of 0.8—4.0μg for HuperzineA (r=0.99964). The average recovery (n=5) was 100.098％for HuperzineA (RSD=1.28％).Compareing extraction effect of five acids, choosing hydrochloric acid as optimization solvent. Ascertain marinate the 1％of hydrochloric acid was selected as the most effective solvent, scale of solid and liquid is 1:30, time is 30h, recovery of Hup A up to 90.36％by orthogonal experimental design.Using active carbon concentrate and enrichment extraction of Hup A. After concentrate and enrichment extraction of HUp A, purity of HupA reach 2.7％. Base on it, using silica gel, alumina, C18, CN pure Hup A, its purity reach 92.7％, recovery reach 93.12％.Innovation is that establish the analyse method of Hup A, research on new technics from extraction to pure. Compared to traditional method, this technics improve recovery and rurity, offer good foundation for producing abundant Hup A. |
