OBJECTIVE: With the model of experimental autoimmune premature ovarian failure mice,we try to see the therapeutical effects of Bu-shen-huo-xue Fang on mice with experimental APOF , and study on the primary mechanism through which Bu-shen-huo-xue Fang improve the ovarian function.METHOD: Seventy-five healthy mice, except 15 for the normal group (N), we immuned all the other 60 with zona pellucida 3 pepitide as antigen. After models were built, we divided all the 60 mice into following four groups: model group (M), estradiol group (E), prednison group (P), and Bu-shen-huo-xue Fang group (Z). And then began to feed the normal group (N) and model group (M) with distilled water, while all the other three groups were fed with Bu-shen-huo-xue Fang liquid or western medicine turbid liquid respectively, 0.5 ml for each mouse every day. Six continuous days with one retired,till four weeks on totol.Then we picked off their eyeballs to get venous blood, also picked up both sides of ovaries.The items were examined with ELISA as follow:serum E2, FSH, and AzpAb. After being weighted, their right ovaries, slicing through stuffing regular paraffin,then for IHC for us to observe the growth o the follicles of different grades as well as the expression of the FSH receptor protein; the left ovaries were examined by RT-PCR in order to see the contents of FSHRmRNA. RESULTS :(1). Compared with the normal group, models have a higher disorder rate of estrus cycle.(P<0.0045)much lower level of E2.(P<0.01)much higher level of FSH.and AzpAb(both P<0.01)while no difference in content of FSHRmRAN (P>0.05).(2)Compared western medicine groups with the model group, estradiol group have a lower disorder rate of estrus cycle(P<0.0045); higher level of E2.(P<0.05)and a much more lower level of FSH(P<0.01),a higher level.of FSHRmRAN(P<0.05); prednison group have a higher level of E2.(P<0.01)while FSHand AzpAb both getting lower obviously(P<0.01).(3) Compared Bu-shen-huo-xue Fang group(Z) with the model group: Z group have a have a lower disorder rate of estrus cycl(eP<0.0045); higher level of E2(.P<0.01);both the level of FSH and AzpAb were lower(P<0.05); a higher level.of FSHRmRAN(P<0.01)(4) Compared Bu-shen-huo-xue Fang group(Z) with the western medicine groups: we find that in Z group AzpAb is lower than that of estradiol group(P<0.05); a higher level.of FSHRmRAN than estradiol group ,while even higher than prednison group(P<0.01) there were no obvious differences between group E,P and Z in other items inferred above.CONCLUSION: 1.Bu-shen-huo-xue Fang can play an effective role on mice with APOF.2. Bu-shen-huo-xue Fang may increase the level of E2 and decrease FSH; inhibit the reaction caused by antigen-antibody complex; increase the content of FSHRmRNA. thus to improve the ovarian function.
