Transdermal delivery of drugs is with advantages of convenience, sustainalbility and controllability compared with traditional administration including the oral administration and hypodermical injection.It provides a new administration method for desease therapy via the research and application of trasdermal delivery in recent years.TD1 is a short peptide with 11 aa: ACSSSPSKHCG.It has been reported to have the potential ability to facilitate transdermal delivery of macromolecules such as insulin through intact skin to reach systemic circulation to therapy desease by coad-administration.Here we constructed TD1-EGF plasmid and purified the fusion protein through molecular biology methods.We found that the TD1-EGF has the similar cell bio-activity compared with the general EGF such as promoting cell growth,migration and stimulating the phosphorylation of ERK1/2.Meanwile, TD1 can promote EGF to penetrate intact skin more easily.The function of TD1-EGF was time and dose dependent,and need normal body temperature..It may be the result of the interaction between TD1 and skin but not protein.Our research of TD1-EGF in cell bio-activity and transdermal delivery ability provides the rationale of TD1-EGF's application in transdermal drug delivery,wound repair and cosmetic.
