Study On The Expression Pattern Of Histone Demethylases In CD4+T Cells Of MRL/1pr Lupus-like Mice

To investigate the expression of histone demethylases in CD4+ T cells of MRL/lpr lupus-like mice.CD4+T cells was isolated from MRL/lpr lupus mice and MRL/MPJ wild-type mice. Real-time quantitative RT-PCR was used to detect the expression of thirteen members of Histone demethylases (HDMs)-LSD1, JMJD3, UTX, JHDM1, JHDM2A, JMJD2A, JMJD2B, JMJD2C, JMJD2D, JARID1A, JARID1B, JARI1C and JARID1D. Western blot were used to detect the protein levels of JHDM1, JHDM2A, JMJD3 and JMJD2B.The results showed that mRNA levels of JMJD2A, JMJD2B, JMJD2D, JARID1C, JARID1D and JHDM1 were significantly decreased in CD4+ T cells from MRL /lpr mice compared with MRL/MPJ mice (P= 0.026, P= 0.006, P= 0.003, P= 0.002, P= 0.019, P= 0.028 respectively). JHDM2A and JMJD3 mRNA were significantly increased in CD4+ T cells from MRL /lpr mice compared with MRL/MPJ mice (P=0.049, P=0.049).However, there was no difference for JMJD2C, JARID1A, JARID1B, UTX and LSD1 mRNA between MRL /lpr mice and MRL/MPJ mice (P>0.05).Furthermore, the results from Western blot showed that the protein levels of JMJD2B and JHDM1 were down-regulated in lupus mice compared to the wild-type mice (P=0.050, P=0.041). In contrast, the protein levels of JMJD3 and JHDM2A were increased in lupus mouse, among which the difference of JMJD3 is significant (P=0.036), the JHDM2A is different without significance (P=0.161).The results of the present work implied the aberrant expression of histone demethylases in lupus mice.
