ObjectivesThis study investigated the effects of intravenous administration of Human Umbilical Cord Blood Cells on inflammatory responses and tumor necrosis factor-αexpression after myocardial infarction in rabbits and further analyzed the mechanism of stem cells for myocardial infarction.Methods45 Chinese rabbits were divided into three groups randomly:Cells transplantation group with 15 rabbits were intravenously administrated with HUCBC labeled with bromodexyuridine(BrdU) at 24h after myocardial infarction; Control group with 15 rabbits were intravenously administrated with saline at 24h after myocardial infarction. the sham operation group with 15 rabbits only not receiving ligation of coronary artery. Cardiac funtions were performed by echocardiography at 1st week,2nd week and 4th week after transplantation. Patho-histological appearance of the myocardium was observed by microscope. The BrdU postive cells and the expression of TNF-αand TNF-αmRNA in the myocardium were examined by immunohistochemical method and RT-PCR Results①Compared to control group,transplantation of HUCBCs improved left ventricular function indexs such as LVFS,LVEF at all 3 examinations;②pathological observations showed compared to the transplanted group, the number of inflammatory cells was significantly higher at the infarct bordery in control group;imunohistochemistry demonstrated that transplanted cells labeled with BrdU were found in the myocardial infarction area in the transplanted group.③Compared to control group, the expressions of TNF-αand TNF-αmRNA were reduced in myocardial infarction area in the transplanted group.ConclusionsIntravenous administration of HUCBCs obviously improved the heart fuction and inhibited the inflammatory responses and expression of TNF-αin the peri-infarcted area after myocardial Infarction. The results suggested that inhibition of the inflammatory responses might be one of main mechanisms for stem cell transplatation for myocardial infarction.
