Objective To investigate the expression and significance of Tenascin-C and CD34 in renal clear cell carcinoma using an immunohistochemical S-P method. The relationship between these proteins expression and pathobiology behavior was analysed and the correlation of these proteins was studied.Material and Methods (1) Immunohistochemical S-P method was used on 56 renal clear cell carcinoma and 15 normal renal tissue for the detection of Tenascin-C and CD34 expression. The relationship between these proteins expression and pathobiology behavior was analyzed. (2) The data were analyzed by software SPSS11.0,χ2-test, exact probability-test and group t test were used to compare difference between groups and Spearman rank correlation was used to analyze the correlation between the two proteins.α=0.05 was considered as level of a test.Results Tenascin-C expression was located in the stroma of renal clear cell carcinoma. The proportion of positive TN-C expression was 53.57% (30/56). The positive rate in well-differentiated,moderately differentiated and poorly differentiated were 25.00% (4/16),47.06%(8/17) and 78.26%(18/23), respectively, There were significant difference(P<0.05) when comparison was made between each of groups. The positive rate of metastasis RCCC was 100%(7/7), while The positive rate of non-metastasis RCCC was 41.07%(23/49), and there were significant difference(P<0.05) when comparison was made between them. TN-C was not detected in normal renal tissues. CD34 showed limited expression in normal renal tissues, the counts number is 11.50+7.11. In renal clear cell carcinoma tissues, the counts number in well-differentiated,moderately differentiated and poorly differentiated were 34.43±6.35,41.76±7.42,52.89±5.51, respectively, there were significant difference(P<0.05) when comparison was made between each of groups. The counts number in poorly differentiated tissues was higher than that in well-differentiated carcinoma tissues, there were significancet difference between them (P<0.05). The counts number of CD34 in metastasis and non-metastasis renal clear cell carcinoma were 57.36±6.76 and 39.43±7.00, respectively. There was significant difference(P<0.05). TN-C expressed in renal clear cell carcinoma was related with MVD positively, r=0.966, P<0.05.Conclusions The expression of TN-C is positively corresponded with the pathologic grade and metastasis. And the expression of CD34 is also positively corresponded with the pathologic grade and metastasis. CD34 is positively corresponded with TN-C. (1) Over expression of TN-C and CD34 were involved in RCCC genesis and the correlation between the expression of TN-C and CD34 and some clinic pathologic factors such as differentiation and metastasis suggested that these proteins were involved not only in RCCC genesis but also in tumor metastasis. (2)TN-C was possiblely an adhesion inhibitor and may promote the growth of epitheliums in the progress of RCCC genesis. CD34 is positively corresponded with TN-C, TN-C plays an importannt role in the RCCC genesis and growth by up-regulating MVD in RCCC possiblely. (3) The united detection of TN-C and MVD can predict the ability of invasion and metastasis of Renal clear cell carcinoma and supply evidence for clinical therapy and judgment of prognosis.
