Objectives:To observe the effect of nano-hydroxyapatite (nano-HA) and platelet-rich plasma (PRP)compound on bone defect and bone construction arounding the rabbit tibia titanium with nanometer-hydroxyapatite(nano-HA) alone or with platelet-rich plasma(PRP).Materials and Methods:24 healthy New Zealand white rabbits were divided into 3 groups (n=8) with self-control method, We implanted a titanium metal body in each one tibia upside 1/3 and 2 cm from the knee and established bone defect at oneside:the left side as experimental group,filled with nano-HA PRP complex; the right side as the control group, filled with nano-HA. All the animals were killed after 2,4,6 weeks respectively and we observed the healing situation of rabbit tibial bone by X-ray, histological staining and acqusited the slice image of new bone defects by computer image analysis system acquisition,and account for the percentage of new born area in the visual field and caculated the metal body bone mineral density and to analysize.Results:â‘ All rabbits were normal in diet and activity after surgical operation, and no animals died or wound infection. The wound healed well and the suture was off 1 week later.â‘¡Gross obsrvation: the top of titanium metal body of both group were covered by parenchyma and experimental group was density than control group; the bone defect of both group were concave.â‘¢specimen X films showed no efluxion and bone defects could be seen after 2 weeks in the visible 4 weeks after surgery, the defect has diappeared after 6 weeks in the control group. Compared with control group, the bone density gray value in experimental has significant difference (p<0.05).â‘£specimens 2,4,6 weeks after histological analysis showed new bone area of the experimental group was significantly different (p<0.05)compared with the control group.Conclusions:(1)Nano-HA alone metal body for filling bone defects showed a certain osteogenic effect;(2)PRP/nano-HA compound showed bone conduction and bone induction ability, which speed up the integration of bone and repair bone defects experiments compared with group nano-HA.
