Objective:To study the expression of E-cad and matrix metalloproteinases 7and 9 in normal ovaries, benign ovarian tumors and epithelial ovarian carcinomas, as well as the correlations with several clinixopathologix and demonstrable prognosticparameters.Method:The representative paraffin sections were selected and 30 specimens of normal ovaries,30 specimens of benign ovarian tumors and 52 specimens of epithelial ovarian carcinomas were tested by immunohistochemical technical with antibody against E-cad,MMP7and MMP9 was used to detect vascular endothelial cell.Results:1. E-cad staining was located on the tumour cell membrane and a few cases'cytoplasmical..E-cad expression was absent in normal ovarian surface epithelium. In epithelial ovarian carcinomas, E-cad expression was different from each ther, compared with staining of benign tumors, E-cad expression in epithelial ovarian carcinoma was apparently decreased, which was statistically significant (P<0.05)2. The expression of MMP-7 and MMP-9 in epithelial ovarian carcinomas were significantly higher than them in benign ovarian tumors and normal ovaries(X27= 30.793, P<0.05; X29=13.781, P<0.05),also MMP-7 and MMP-9 were strong express in epithelial ovarian carcinomas. The difference between benign and malignant tumors was significant(t=9.713,P<0.05;t=2.233,P<0.05).3. Higher expression in mucinous cancer than in other type of ovarian cancers; A reverse correlation were found in the expression of E-cad and the histological grade(r=-0.551, P<0.05); As to clinical stage, E-cad expression was significantly higher inⅠ~Ⅱstage than inⅢ~Ⅳstage(t=2.151, P<0.05); The expression of E-cad in patients of ascites without tumor cells was significantly higher than its in the patients of ascites with tumor cells(t=2.246, P<0.05).4. In epithelial ovarian carcinomas,MMP7,MMP9 staining was located in the cyto-plast and on the cell membrane of both endothelial cells and tumor; In epithelisl ovarian carcinomas, the expression of MMP7,MMP9 was significantly different with different clinical stage and histological grade (F=3.662, P<0.05; F=4.887, P<0.05;F=11.353,P<0.05;F=4.334,P<0.05 respectively). It's no significantly with the expression of MMP7 and MMP9 with different histological in epithelial ovar- ian carcinomas. The patients with higher expression of MMP7,MMP9 had unfavourable prognosis than them with lower or nrgative expression (Log-rank,P=0.0286). The expression of MMP7 and MMP9 in patients of ascites with tumer cells was significantly higer than their in the patients of ascites without tumor cells(t=3.623,P<0.05;t=3.520,P<0.05).5. The reverse correlations between E-cad and MMP7,E-cad and MMP9 were significantly related in epithelial ovarian carcinomas; and the correlations between MMP7 and MMP9 was divorce (r=-0.285, P<0.05; r=-0.394, P<0.05; r=0.921,P<0.05).6. It's better survival prognosis in the strive masculine expression of E-cad than the minus and feebleness cases, which was no statistically significant(P>0.05);but for MMP7 and MMP9,It's less survival prognosin in the strive masculine expression of them than the minus and feebleness cases, which was statistically significant (P<0.05).Conclusion:In epithelial ovarian carcinomas, E-cad was higher expressed in highly differentiation and early stage tumors, but, the expression of MMP7 and MMP9 are significantly different in either different clinical stage or different histological grade. The expression of E-cad,MMP7 and MMP9 are significantly correlated with ascites, indicating MMP7 and MMP9 have the importance of accelerate angiogenesis, and it is E-cad The expression of MMP7 and MMP9 are significantly correlated with E-cad, indicating they are important factors of occur and development in epithelial ovarian carcinomas, E-cad,MMP7 and MMP9 are prognostic factors in epithelial ovarian carcinomas.
