Objective To establish rat unstable plaque model by high-fat diet (PTU added) and intraperitoneal injection of VitD3 or ovalbumin.Methods 30 male Wistar rats were randomly divided into 3 groups. Group A: high fat diet (PTU added) + VitD3 intraperitoneal injection;Group B:high fat diet (PTU added) + VitD3 and ovalbumin (OVA) intraperitoneal injection. Group C was control group with normal diet and saline intraperitoneal injection. Before the intervention. tail venous blood of the rats was drawn and tested for blood biochemical parameters and inflammatory factors. At 8 week, sodium pentobarbital was intraperitoneally injected in a dose of 30 milligram per kilogram of rats, weight to anesthetize the rats. A blood was drawn from the rats,heart and tested for blood biochemical indexes and inflammatory factors. Entire aortas were taken and used for conventional morphology and immunohistochemical tests.Results 1.The rats of Group B and Group A have already devloped obvious hyperlipidemia. The total cholesterol (TC), triglyceride (TG) and low density lipoprotein (LDL) of group B and A were significantly higher than that of the control group while the high density lipoprotein (HDL) of B and A group were significantly lower than that of the control group.2. The interleukin -6 (IL-6) and oxidized low density lipoprotein (ox-LDL) of Group A and B were significantly higher than that of the control group.3. Comparing with those of Group A and the control group, the Ballooning degeneration in the livers of Group B obviously increased, the aortic atherosclerotic plaques of Group B were more obvious in HE staining test, and the mast cells of Group B increased obviously in immunohistochemical test.4. Comparing with those of the control group, the number of mast cells in the shoulder area of group B and A were obviously increased.Conclusion high-fat diet (PTU added) and intraperitoneal injection of VitD3 and ovalbumin could establish the Wistar rat model of unstbale atherosclerotic plaques.
