Objective: Canthium horridum Bl. is one of the four Canthium species growing in China, which is a frutex distributing mainly in Guangdong, Guangxi and Yunnan provinces, P.R.China. The genus is known for its folkloric applications against a number of ailments such as cough, diabetes, and hypertension. Some plants of this genus are used as antipyretic, anti-inflammatory agent in folk medicine. In order to take comprehensive utilization, it is very useful for studies on chemical constituents and antimicrobial activities of Canthium horridum Bl..Method: The air dried C. horridum Bl. are crushed, and then extracted by 75% ethanol (in volume fraction). After evaporation under reduced pressure a dark green ethanol extract was obtained. The extract was mixed with water, extracted by petroleum ether, chloroform, ethyl acetate and n-BuOH, respectively. Then take a test on anti-bacterial activity of different fractions. The petroleum ether, the chloroform, the ethyl acetate and the n-butanol fractions were respectively subjected to column chromatography over macroporous resin, normal and reversed-phase silica gel, sephadex LH-20, preparation thin-layer chromatography and recrystallization repeatedly to obtain pure compounds.Results: Bioassay-guided isolation studies on the stem extract of Canthium horridum Bl. led to the isolation of twenty compounds: Syringaresinol(1), Quercetin(2), Scoparone(3), Scopoletin(4), Fraxidin(5), Sinapic aldehyde(6), 3-Methoxy-4-hydroxy-trans-cinnamaldehyde (7), Coniferyl alcohol(8), p-hydroxybenzoic acid(9), Vanillic acid(10), Syringic acid(11), Syringaldehyde (12), Di-isobutyl phthalate(13), Vanillic acid 4-O-β-D-glucopyranoside(14), 3-(1-C-β-D-glucopyranosyl)-2,6-dihydroxy-5-methoxybenzoic acid(15), Mannitol(16),β-daucosterol(17),β-sitosterol(18), Nonadecanoic acid(19), Dibutyl phthalate(20). Compounds (1), (2), (5), (7), (8), (9), (12)-(16), (19), (20)were reported for the first time from this genus. The antimicrobial activities of compound (1), (3), (4), (7), (11), (14) were studied, the results were as follows: Compound (1) had high inhibitory activities against Escherichia coli, Bacillus subtili and Staphylococcus aureus. Compound (3) and (7) had medium inhibitory activities against these three bacterias. Compound (14) had low inhibitory activities against E.coli and B.subtili. All the compounds had no inhibitory activities against Aspergillus niger.
