The Effects Of Fuyuan Capsule On Tumor Factor–alpha And Vascular Endothelial Growth Factor In Cartiage Cells Of Experimental Osteoporosis Rats

OBJECTIVE:To observe the effects of FuYuan capsule on TNF-αand VEGF in osteoarthritis,and investigate the mechanism about the reparation and treatment of FuYuan capsule.Methods:Fifty-six rats , OA models were induced by Hulth's technique,were randomly divided into five groups: normal group with 6 rats,model contrast group with 10 rats,tetracycline group with 10 rats,FuYuan capsule low,moderate and high does group with 10 rats.The normal group and model contrast group were fed with 0.9 N.S,tetracycline group were fed with tetracycline and otherswith FuYuan capsule.After 10 weeks, the rats were sacrificed,and then rat tibiaarticular cartilage were removed. Sections of the articular cartilage were stainedwith hematoxylin—eosin.The cartilage histological characteristical were observed according to the method of Mankin.Immunohistochemical staining was performed to investigate the expression of TNF-αand VEGF.Articular cartilage were observed with microscopy and image analysis method was used to measure the expression intensity of TNF-αand VEGFResults:There were significant difference in Mankin score among normal group and other groups(P<0.05). Immunohistochemical staining indicated that the expression intensity of TNF-αand VEGF among normal group and experimental groups were significantly different (P<0.05).Conclusion:FuYuan capsule can protect the articular cartilage through suppressing the TNF-αand VEGF expression in chondrocyte ,with suppre- ssing inflammation and andangiogenesis.
