Objective To observe the expression of suppressors of cytokine signaling one and three (SOCS1/3) mRNA, the change of Help T cell one (Th1) and Help T cell two (Th2) and CD4+CD25+Regulatory T cells (CD4+CD25+Treg) in peripheral blood T lymphocyte cell of children with Kawasaki disease(KD), and to discuss the immune mechanism of KD.Methods 10 healthy children,20 children with pneumonia and 20 children with KD were researched. The SOCS1/3mRNA levels and the expression of IFN-γ, IL-4 and CD4+CD25+Treg of T lymphocyte cells in peripheral blood were analyzed by RT-PCR technique or FACS analysis, respectively.Results The expression of SOCS1/3mRNA were significantly decreased in patients with KD in acute stage compared with those of healthy children and children with pneumonia (P<0.05). The IFN-γand IL-4 levels of T lymphocyte cells in patients with KD in acute stage were significantly decreased compared with those of healthy children and children with pneumonia(P<0.05), and the levels of CD4+CD25+Treg also decreased obviously compared with those of healthy children(P<0.01), but no significant differences were ocurred compared with those of children with pneumonia(P>0.05).Conclusion The percentage of peripheral blood CD4+CD25+Treg decreased obviously in acute stage of KD. The levels of IFN-γand IL-4 produced by T lymphocyte cells and the levels of SOCS1/3 mRNA expressed by T lymphocyte cells of peripheral blood in KD children in acute stage decreased significantly.
