| Objective:To use controllable mechanical model of complete intestinal obstruction, to observe the intestinal mucosal barrier transform and influence of traditional Chinese medicine Sijunzi decoction on it.Methods:The healthy rabbits with body weight of 2.5-3kg were divided into 6 groups randomly, including the sham operation group(SO),ileus of 48 model group(I48),natural recovery 48h group(S48),Chinese medicine treatment of 48h group (T48), natural recovery 96h group (S96),Chinese medicine treatment of 96h group (T96), each group 10. The lifting of an animal model of obstruction after 48h,96h. Making serum D-lactate,serum intestinal fatty acid-binding protein, serum diamine oxidase,organization of intestinal trefoil factor,intestinal mucin, intestinal mucosa goblet cell morphology and Chiu's histopathological evaluation score detection and observation,respectively.Result:①The pathophysiological changes of small intestine of I48 group was in line with the typical small intestinal obstruction,while the pathophysiological changes after relieving obstruction also was in line with the changes of the typical relieving obstruction.②The experiments showed that small intestinal mucosal damages was gradually recovered,Chiu's score gradually was decreased, especially in Chinese medicine the treatment group was significantly higher than the natural recovery group (P<0.05). the number of goblet cells of small intestine epithelium was gradually decline after the lifting of the obstruction, the significant differences have emerged 48h later,96h returned to normal level, traditional Chinese medicine treatment group was more effectively. Serum D-lactate,serum intestinal fatty acid-binding protein, serum diamine oxidase levels gradually returned to normal levels, the Chinese Medicines Board was earlier than the natural recovery group (P<0.05).Also found in traditional Chinese medicine Sijunzi decoction can increase the content of organization of intestinal trefoil factor,intestinal mucin higher than the natural recovery group.Conclusion:①The controllable mechanical complete intestinal obstruction animal model can be established successfully by the method.②Sijunzi decoction can promote intestinal mucosal barrier of the repair process after the lifting of intestinal obstruction, can accelerate the performance of their intestinal intraepithelial goblet cells of the intestinal epithelium of the completion of the reconstruction progress, can increase the content of organization of intestinal trefoil factor,intestinal mucin and promote the integrity of intestinal epithelial repair. The results showed the role of Sijunzi decoction may be the promotion of intestinal crypt stem cell proliferation and goblet cell secretion, improve the intestinal mucosal microcirculation, so as to achieve the integrity of epithelial repair. Thus, reducing the permeability of intestinal mucosal barrier and the incidence of intestinal infection effectively,as to the dialectical application of sijunzi Decoction protect the intestinal mucosal barrier after the lifting of intestinal obstruction to provide some experimental evidence in clinic. |
