Because of the second-order advantage, which can provide quantitative information of analytes even in the presence of interferences, three-way second-order calibrations have been getting more and more popular. During the past three years, aiming at the actual problem that it is difficult to directly analyze pharmaceutical compounds in human plasma or urine, the author investigate the application of using second-order instruments coupled with trilinear decomposition algorithms on the determination of the concentration of pharmaceutical compounds in human plasma which include:1. Determination of Penciclovir and Famciclovir in plasma coupled with parallel factor analysis, PARAFAC. Famciclovir is an antiviral drug. Its metabolite in human plasma is Penciclovir which has the activity and acetic acid which has no toxicity. However, drug overdose will cause side effects on human body's nervous system and digestive system. Besides, Kidney or liver exhaustion will have impact on the process of translation famciclovir to Penciclovir. In this chapter, we study the determination of Penciclovir and Famciclovir in human plasma by PARAFAC coupled with excitation-emission matrices (EEMs).2. Determination of Procaine hydrochloride and Tetracaine hydrochloride by EEMs coupled with APTLD. Both procaine Hydrochloride and tetracaine Hydrochloride belong to aromatic acid salts of local anesthetics, and they are used toLocal anesthesia in clinical medicine. If low dose is used, the anesthesia will not obvious and, thereby, bring pain for patients. However, if more Dose employed, it may lead dangerous to patients. Hence, it is important to detect the concentration of these compounds in human plasma. In this chapter, we determinate procaine Hydrochloride and tetracaine Hydrochloride by APTLD coupled with EEMs at pH=7.0.3. Quantitative analysis of Atenolol in human plasma by combiningPARAFAC with EEMs. Atenolol isβ1 adrenergic receptor blocker. It is mainly used for hypertension, angina, etc. Unfortunately, this medicine has serious side effects, such as hypotension and bradycardia. So it is very useful to monitor its residue in human plasma to provide a guideline in clinical medicine. Fast detection of Atenolol in human is studied by EEMs coupled with PARAFAC.4. The concentration of Hydrochlorothiazide was determined by HPLC-DAD coupled with ATLD. Hydrochlorothiazide is a diuretics, and mainly used for cardiogenic edema, liver edema, iatrogenic edema and renal diseases in clinical. However, overdose may cause low potassium blood high blood uric acid, hyperglycemia and so on. The traditional chromatography surfer the disadvantage such as need critical separate condition, besides, the condition cannot be used for other system directly. In chapter 5, we investigate the determination of hydrochlorothiazide in plasma by HPLC-DAD coupled with ATLD in the presence of overlapped peaks.
