The Pharmacological Effects Of Triptolide In Adjuvant Arthritis Model Affected By Acupuncture

Objective:(1)establishing the adjuvant arthritis (AA) rat model to study acupuncture improved their symptoms.(1)doing the research on the serum concentrations of triptolide in the adjuvant arthritis (AA) rats after acupuncture therapy,and disscuss the triptolide metabolism in the biological model,to provide a reference for the future experimental research and clinical treatment.Methods:SD rats were modeling,divided into five groups:the rats injected with normal saline as a control group,the injection of complete Freund's adjuvant (CFA) in rats after inflammation as the AA rats and randomly divided into model group,acupuncture group,drug group,acupuncture and drug group.(1) observing the changes of body weight of each group,joint swelling,joint pain score among the groups.(2) using high performance liquid chromatography (HPLC) to Hedera CN column (5um* 200mm* 4.6mm) as the stationary phase to 60% water and 40% acetonitrile as mobile phase,flow rate:1.0ml/min;Column temperature:40℃,UV detection wavelength of 217～221nm. Taking he rat plasma,adding a certain volume of ethyl acetate,vortex mixing and centrifuged,imbibing the supernatant and clear part by a certain percentage. The supernatant was placed in 60℃apparatus volatilized nitrogen evaporated,and its residue was dissolved in mobile phase,injected in analysis meter after filtered. Chosing different modeling groups of AA in rats after intravenous injection of a single dose of triptolide;taking blood sampling by tail Department at different time points,analyzing pharmacokinetics parameters of drug in blood and disscussing the drug in the metabolism of the body.Results:(1)after treatment weight,joint swelling,joint pain score flexion of AA rats was significantly higher than normal rats. (2) AA triptolide in rat plasma and the minimum limit for concentration detection was 0.01μg/ml. The extraction recovery of plasma samples gose up to 85.9%. Day and day RSD were less than 15% difference. In the biological samples the concentration of triptolide get a good linear relationship with peak area.Conclusion:This study established the AA rat model,using acupuncture,as a method of treatment for RA,and made a preliminary assessment with its efficacy and comparison. And use HPLC method for the determination of AA rats serum concentrations of triptolide. The method is sensitive,accurate, reproducible,high reliability,simple and easy to promote,to meet the demand of drug in AA rats in vivo Pharmacokinetics studies.
