| Abstract:Objective:To investigate the expression and significance of tight conjunction proteins claudin-3 in meningioma with meningioma genesis,angiogenesis and the relationship between the expression of tight conjunction proteins claudin-3 in meningioma with clinicopathology of meningioma. To uncover the relationship between meningioma genesis and angiogenesis with the expression of claudin-3 and expect finding new ways to diagonose and treat meningioma. Methods:The expression of tight conjunction proteins claudin-3 levels were detected by immunohisto-chemical (SABC) technique in 63 meningioma tissues and 10 human normal cerebral maters. CD105 endothelial antigen were used to label the proliferative activity and new growth microvessel, MVD in meningioma and human normal cerebral maters were calculated. Collect the clinical and pathological data and analyze them statistically with SPSS software.Results:①63 cases of meningioma tissues, claudin-3 was weakly positive (+) in 17 cases, positive (++) in 24 cases and strongly positive (+++) in 22 cases.10 cases of normal meningeal tissue claudin-3 strong positive(+++)。Expression of claudin-3 was significantly lower than the normal dura mater, the difference was significant (P=0.00<0.05).②10 cases of normal meningeal tissue MVD was 3.6±1.05,63 cases of meningioma in the mean MVD was 38.44±10.74, the difference between the two was significant (P=0.017<0.05).③In meningioma tissues, the expression of claudin-3 was weak positive (+) cases, MVD was 56.88±11.17, the expression of claudin-3 was positive (++)cases, MVD was 35.43±4.14, the expression of claudin-3 was strong positive (+++)cases, MVD was 28.30±3.94; the expression level of claudin-3 was negatively correlated with MVD (rs=-0.613, P=0.00<0.05).④In 63 cases of meningioma tissues, claudin-3 positive levels of different gender (χ2=1.107, P=0.547>0.05), different age (x2=2.964, P=0.225> 0.05), different tumor sites (χ2=5.989, P=0.829> 0.05), different tumor shapes (χ2= 4.015, P= 0.710> 0.05)was no significant difference, and in different tumor grade (χ2=45.213, P=0.00<0.05), clinical condition (χ2=24.6417, P=0.000<0.05), tumor invasion (χ2= 7.965, P=0.017<0.05),the expression was significantly different。Conclusion:①Expression of claudin-3 in meningioma was down, and with the increase of lower-level cases, stronger positive expression in normal meninges.②Claudin-3 in meningioma tissues and MVD was negatively correlated, with the increase of claudin-3 expression decreased microvessel density also increased. above suggest claudin-3 and tumor cell proliferation and angiogenesis, clinical pathology is closely related, claudin-3 predicted meningioma may be an important indicator of biological behavior. deepen research the adjustion principle of the expression of claudin-3, we may find a new way to diagnose and cure meningioma. |
