Scutellaria barbata D.Don is a wild perennial plant endemic to southern China, which has been widely used for medicinal purposes, such as in inflammation, hepatitis, ostemyelitis, gynecological, lung and rectal tumors in traditional Chinese medicine. On the other hand, Oldenlandia diffusa (Willd.) Roxb. is well-known folk medicine in southern China and has been used for the treatment of hepatitis, tonsillitis, sore throat, appendicitis, urethral infection and malignant tumors of the liver, lung and stomach. Frequently, these two traditional Chinese medicine plants were used together, named "herb pair" in traditional Chinese medicine, to get mutual enhancement or mutual assistance for the treatment of cancer and tumor in the clinical practice.Chemical constituents of traditional Chinese medicine were the basis of exerting its medicinal efficacy, so this paper aimed at studying the substantial basis of the extracts from the herb pair of Scutellaria barbata D.Don and Oldenlandia diffusa (Willd.) Roxb. based on the chemical constituents. Its research thoughts followed the combination of chemical separation and activity screening, medicinal efficacy was selected as the counted unit and bioactivity-guided chromatographic separation was used to obtain the active part or effective constituents. According to medicinal efficacy and clinical effect on a certain disease, the rapid screening model was established and these models were used to corroborate active part or effective constituents. Furthermore, the qualitative identification of the effective constituents was made by HPLC-MS and UV. Based on these data, the paper inferred the source of effective constituents, checked whether it could produce new constituent, analyzed the connection between medicinal efficacy and change of quality and quantity, and elucidated the compatible regularity of herb pair and therapeutic mechanism to a certain degree, finally.In the first chapter, the study progress in medicinal herbs differentiation, chemical constituents, pharmacology effect and clinical application of Scutellaria barbata D.Don and Oldenlandia diffusa Willd. was summarized. The pharmacology effect and clinical application of herb pair about tumour terapy were introduced and the connection between tumourigenesis and excess of radicals was discussed. Finally, the research object, research significance and research content were described in detail.In the second chapter, the extract (F1) of herb pair from Scutellaria barbata D.Don and Oldenlandia diffusa Willd. was enriched by macroporous resin and each effective part was prepared from macroporous resin enrichment(F2) by polyamide column chromatography. Total phenolics and flavonoids content in the extracts were determined.In the third chapter, the antioxidant and antibacterial activity of F1,F2 and all effective parts were evaluated by activity screening models. The result showed that the antioxidant and antibacterial activity of extracts were significantly related to their composition and content.In the fourth chapter, by mean of the HPLC-MS and UV, the effective constituents of F2-10 and F2-20 were identified and their chemical characters were determined. Furthermore, the effective constituents were prepared and their activities were studied combining with active screening models. The result showed that the fraction F2-20 mainly contains three quercetin-3-o-glycoside derivatives, that is Quercetin-3-O-sambubioside(J),quercetin-3-O-(2-O-.-D-glucopyranosyl)-.-D-galact opyranoside(E),quercetin-3-O-[2-O-(6-O-E-feruloyl)-.-D-glucopyranosyl]-.-D-galac topyranoside(Y). Using the quercetin (H) as the positive control, the antioxidant capacity of quercetin and its derivatives was arranged as follows:H> Y>E>J. The fraction F2-10 mainly consisted of p-coumaroyl scandoside methyl ester, and the antibacterial activity of this compound was weaker on Bacillus subtilis. By analyzing the experimental data, it is found that the antibacterial activity of F2-10 was not provided by one or some effective constituents, while was due to the results of interaction of all constituents.
