The theory of"man,as a microcosm,is connected with the macrocosm"is an important component in the Chinese medical theoretical system, which plays an important guiding role in the prevention, diagnosis and treatment of diseases. The impact of climate changes of four seasons on human organs'functions is the core of the theory and the core of Chinese medical clinical treatment. From the perspective of"lung disseminated air gas, wei-energy has a defensive function",this study does research about the impact of seasonal changes on the anti-infection cytokines of rat's respiratory system, exploring the rules of changes in the anti-infection cytokines resulted in the seasonal changes and the immunological basis of respiratory disease causes, so as to provide theoretical basis and guidance for the modernization studies clinical preventions of the theory of"man,as a microcosm,is connected with the macrocosm,"and for the treatment of infectious respiratory diseases.Objective: This experimental study is theoretically based on the statement about the impact of seasonal changes on human organ functions in the theory of"man,as a microcosm,is connected with the macrocosm"in Huangdi Neijing.By abserving the changes in the lung tissue slices of rats, this study explores the law of changes of immunological indicators related to respiratory diseases due to seasonal climate variation so as to provide theoretical basis and experimental evidence for the prevention and diagnosis treatment of seasonal infectious respiratory diseases.Methods: This study selected healthy male SD rats with weight of 180-200g which purchased from Experiemental Animal Center of Hei Bei Medical University 14 days before each solar term.The rats were divided into 8 groups, the Spring Commence, Spring Equinox, Summer Commence, Summer Solstice, Autumn Commence, Autumnal Equinox, Winter Commence, Winter Solstice group.Each group included 8 rats.The rats are fed with regular complete formula feeds under laboratory conditions with room temperature, exposed to natural light, and are free to intake water. The rats are beheaded at 6:00pm on the day of each solar term. 5-10ml of blood from each rat.Interleukin( IL-1β, IL-2 and IL-6) in the serum are detected by the method of Radioimmunoassay.Interferon-γand IL-10 in the serum were detected by Enzyme-Linked immunosorbant assay.At last observe and analyse the variation of the indicators,then sum up the two groups in each season to detect the law of the the variation.Results:1 statistical analysis from the angle of solar term variation:The level of IL-1 in the serum of rat is lower in Spring Commence, Winter Commence,and The Winter Solstice group than other solar terms,while the level of IL-1 in the serum of rat is higher in The Spring Equinox,Summer Commence,Summer Solstice,Autumn Commence and Autumn Equinox than other groups. There is significant statistical difference between the former 3 groups and the later 5 group(sP<0.05).There is no statistical significance among Spring Commence, Winter Commence,and Winter Solstice in the level of IL-1 in the serum of rats. The level of IL-1,IL-2 in the serum of rat is lower in Spring Commence,Winter Commence ,and Winter Solstice group. The level of IL-6in the serum of rat is lower in Spring Commenceand Winter Solstice group. The level ofIFN-γin the serum of rat is lower in Winter Commence ,and Winter Solstice group. Statistical significances from the angle of seasonal variation. The level of IL-6 in the serum of rat is lower in Winter and Spring group. The level of IL-1,IL-2,IFN-γin the serum of rat is lower in Winter group. The above results indicate that anti-infection cytokines of rat's respiratory system in winter and spring are relatively low . The result to some extent explicated why respiratory diseases always occur in Winter and Spring when the body immunological function does not work well,The experimental results provide scientific evidence to reveal the immunological mechanism for the seasonal incidence of respiratory diseases and provide guidance to clinical prevention,diagnosis and treatment of seasonal respiratory diseases.2 Experimental results indicates that anti-infection cytokines of rat's respiratory system has some changes due to the seasonal variation. It further supports the theory that the body will make adaptable adjustments and changes due to the seasons, which is rooted in the doctrine of"correspondence between man and universe".
