| Objective: To investigate the possible correlation and inflammation mechanism between chronic periodontitis (CP)and chronic renal failure(CRF) by establishing chronic periodontitis and chronic renal failure model in SD rats first. Methods: Forty health male SD rats were divided into four groups: A(control group), B(CP group), C(CRF group), D(CP accompany with CRF group), 10 Rats in every group. Established CP model by ligating silk suture and besmeared pathogenic bacterium in rats dental cervix; Established CRF model by 5/6 Nephrectomy. All rats were sacrificed after 8 weeks to detect periodontal index and levels of serum Scr and BUN. The expression of IL-1βand TNF-αand hs-CRP in serum and nephridial tissue were examined by ELESA and immunohistochemistry method. The severity CP and CRF was quantified by histopathology. The data were statistically analyzed. Results: The correlated detection revealed animal models were established successfully. Renal function revealed that Scr and BUN in group Cand D were higher than that in group Aand B(P﹤0.05), and that in group D was higher than group C(P﹤0.05), the differences of Scr and BUN revealed between group A and B were no significant(P>0.05). Histopathology observation revealed that the kidney change of inflammation was observed in group B, the score of PAS and Masson in group Cand D were higher than that in group A and B(P﹤0.01), and group D was higher than group C(P﹤0.05). Obvious inflammation of periodontal tissue was observed in group B and D. AL and mBI in group B and D were higher than that in group A and C(P﹤0.05), and that in group D was higher than that in group B(P﹤0.05). AL in group C was higher than that in group A (P﹤0.05). The level of serum IL-1β,TNF-αand hs-CRP in group A and B and C were higher than that in group A(P﹤0.05), IL-1βin group D was higher than that in group B and C(P﹤0.05), TNF-αand hs-CRP in group D was higher than that in group B(P﹤0.05). The expression of TNF-αand hs-CRP and IL-1βin nephridial tissue were compatible with that levels of serum.2×2 factorial design revealed, there existed interactions between CP and CRF on the numerus of Scr and BUN and AL(P﹤0.05), and the influence of each factor on that were significant(P﹤0.05), no interactions was noted between CP and CRF on IL-1β,TNF-α,hs-CRP(P﹥0.05), but the influence of each factor on that were significant(P﹤0.05). Conclusion: The model can be used to observe the influence between chronic periodontitis and chronic renal failure, there have correlation between chronic periodontitis and chronic renal failure, chronic periodontitis can aggravate chronic renal failure, chronic periodontitis maybe make more serious lesion in renal by inflammation mechanism. |
