Objective:To evaluate the clinical curative effectiveness, the time-effect relationship and the safety of Banxiaxiexin decoction in treating insulin-resistant and polycystic ovary syndrome.Methods:Select 84 women who visited gynecological clinics in China Academy of Traditional Chinese Medical sciences Guang’anmen Hospital in accordance with the inclusive and exclusion criteria and with the diagnosis of IR and PCOS from June 2014 to December 2015. Taking Banxiaxiexin decoction as the basic prescription and modified according to the TCM differentiation for a period of 6 months. The oral glucose tolerance test and insulin release test were performed and the area under glucose and insulin curve were measure before and 3 months,6months after the treatment, as well as hormone levels (FSH, LH, E2,T, PRL), CHO, TG, LDL-C, HDL-C, ALT, Cr, BUN. The menstrual cycle scores, the TCM symptoms scores, height, weight, waist circumference, hip circumference, BMI, WHR were recorded before and 3 months,6months after the treatment. The rate of pregnancy was counted after 3 and 6 months’ treatment.Results:1. Compared with before the treatment, P2h-INS and area under insulin curve were significantly reduced after 3 and 6 months’treatment (P<0.05). What’s more, after 6 months’ treatment, the degree of declining in P2h-INS was particularly significant than that of before the treatment(P<0.01). Compared with before the treatment,1 and 2 hours’post-meal blood glucose and area under blood glucose curve were significantly reduced after 6 months’ treatment (P<0.05). There was significantly difference between 3 and 6 months’ treatment in the reduction of P2h-PG (P<0.05).2. Compared with before the treatment, LH, FSH, T were significantly reduced after 6 months’ treatment (P<0.05) and the degree of declining in T was particularly significant than that of before the treatment(P<0.01). And LH was significantly reduced after 3 months’ treatment (P<0.05). But there was no difference between 3 and 6 months’treatment in the decreasing of LH, FSH and T.3.The cumulative rate of pregnancy during the first 3 months’treatment was 13.21% and during the second 3 months’treatment was 41.30%, having a significant difference (P<0.05).4. Compared with before the treatment, menstrual cycle score was significantly reduced after 3 and 6 months’ treatment (P<0.05), but there was no difference between 3 and 6 months’ treatment.5. Compared with before the treatment, scores of TCM symptoms was significantly reduced after 3 and 6 months’treatment (P<0.05) and the degree of decreasing was particularly significant after 6 months’treatment(P<0.01). But there was no difference between 3 and 6 months’ treatment.6.Compared with before the treatment, there was no difference in ALT, Cr, BUN after 3 and 6 months’ treatment.Conclusion:1. Banxiaxiexin decoction can improve insulin sensitivity and reproductive endocrine status in treating PCOS and IR and there is time-effect relationship.2. Banxiaxiexin decoction can improve clinical symptoms, promote autonomous menses recovery and improve the chances of pregnancy in treating PCOS and IR and there is time-effect relationship. It may relate to the improve of insulin sensitivity and reproductive endocrine status.3. Banxiaxiexin decoction have no impact on hepatic and renal function.
