The studied cyanobacteria isolated from Haematococcus pluvialis was an endophyticcyanobacteria. Preliminary sequencing and electron micrographs showed that this strain is anew species of cyanobacteria, it had different internal structure under differentlight-conditions.Obvious Chloroplast structure is found in HL-algae.In this paper,thepigment compersition in two conditions were studied. It provided a basis for chloroplastorigin by measuring its pigment composition and content.The main results of this study were as follows:The separation and purification experiments of Chlorophyll determined that the bestextraction solvent was100%acetone, which could extracted fully. Selected the paperchromatography method for separating, it developed with Petroleum ether: acetone7:3(V/V), the determine of spectroscopic characteristics further defined the types ofHL-algae:chlorophyll a and chlorophyll b, there was only chlorophyll a in LL-algae. Andcalculated the content.The separation and purification experiments of Carotenoids determined that the bestextraction solvent was100%acetone,it was easier to separate the petroleum ether andacetone when concentrated. Selected the paper chromatography method for separating wasquickly and easy.β-carotene was developed with petroleum ether: acetone7:3(V/V),luteinwas developed with petroleum ether: acetone: isooctane20:2:1.In order to achieve the bestseparation, adding a few drops of acetic acid to adjust the polarity of the solvent. Thedetermine of spectroscopic characteristics further defined the types and cotent. β-caroteneand lutein were found in HL-algae, only β-carotene was contained in LL-algae.The separation and purification experiments of Phycocyanin(PC) determined that thebest extraction agent is0.75M PBS buffer. After settling overnight, using secondary saltingas purification method,first of25%ammonium sulfate, secondary of55%ammoniumsulfate,the precipitate was dissolved with0.01molPBS which was the best. UV absorptionspectra and fluorescence emission spectra of Phycocyanin extract measured afterpurification, further provided the basis for defining and calculating content.And concluded:only C-PC with a high concentration was found in LL-algae, no PC was found in HL-algae.The experience drew that energy transfer mechanism under LL is light-most chlorophylla, β-carotene-chlorophyll a. Energy transfer mechanism under HL is: light-chlorophyll b, carotenoids, the majority chlorophyll a-chlorophyll a. LL-algae has thetypical characteristics of prokaryote from the pigment composition analysis. After changingin light intensity, the composition of HL-algae pigment tend to eukaryotes. We could knowby the endosymbiosis theory Chloroplasts originated fromprokaryotes cyanobacteria whichwas autotrophic photosynthetic. It is a symbiotic. In this experiment the newly discoveredabout the optical system provided a new basis on endogenous theory.
