Objective To study Han population and Canarium album(Lour.)Raeusch from Sichuan Province along Yangtze River in China groups geneticpolymorphism data which from southern Sichuanor luzhou.Methods PCRamplification and polyacrylamide electrophoresis of PAGE with high sensitivesilver staining method, namely Amp-FLP of120Han nationality in Luzhou area,Sichuan unrelated individuals D1S80and D16S539loci were polymorphicanalysis. The improved RAPD analysis was performed in nine samples byprolonging the ramp time from annealing to extension with ten RAPD primers,which were selected randomly to analyze the genetic characteristics. Results Inthe survey of120unrelated individual samples, observed D1S80loci with18alleles, gene frequency distribution in the0.004166667-0.1958333333,heterozygosity was0.7869, the power of discrimination was0.87, exclusionrate of PE was0.8152, the genotype distribution was in accordance withHardy-Weinberg balance rule, whereas observed D16S539loci with10alleles,gene frequency distribution in the0.004166667-0.2, heterozygosity was0.7907,the power of discrimination was0.9416, exclusion rate of PE was0.6164, thegenotype distribution was in accordance with Hardy-Weinberg balance rule.The polymorphism of the nine C. album was86%, and the similarity coefficientranged from0.65to1.00. Among the similarity coefficients of the cultivars, the similarity coefficient of sample No.2,3and6was highest (1.0), while thesimilarity coefficient of sample No.5and7was0.98, which indicated that theyare the same cultivars. The similarity coefficient of sample No.9and4was thelowest (0.65), which indicated that they are the different cultivars. ConclusionBoth D1S80and D16S539gene loci of PCR type have good accuracy andsensitivity. In addition, it is also good genetic markers for forensic scienceapplication and research on individual identification and paternity test, geneticdisease gene linkage analysis;Our study provides a theoretical basis for thebreeding and classification of C. album in Sichuan Province along YangtzeRiver of China by using improved RAPD. Also this study suggests that theimproved RAPD amplification could be an effective technique to analyze thepolymorphism and genetic relationship in different organisms, especially, C.album.
