LiuPao tea is a dark tea manufactured in the Guangxi province, it is believed to have digestive and anti-obesity effect. As a kind of fermented tea, microbial fermentation is not only involved in the processing of the tea, but also has important influence on its special quality and function in health care. But studies on the microbes of LiuPao tea are less and not deep enough so far, its microflora is unclear. This study investigated the fungal colonizations exisiting in LiuPao tea which produced in three factories, in order to fully understand its fungal diversity, identify the predominant fungi to provide theory support for LiuPao tea’s quality, safety and deep development in the future.The research results are as follows:The fungal diversity and community structure in four LiuPao tea samples were studied with high-throughput sequencing technology, and3phyla, seven classes,9orders,6families and8genera of fungi taxa were identified, including Aspergillus、Eurotium、Penicillium、Blastobotrys、Guehomyces、Rasamsonia、 Rossbeevera and Emericella.The relative abundance of Aspergillus and Eurotium were99.85%、90.65%、99.94%and60.37%in the four tea samples respectively, and there are37.69%unclassified information in CY4samples.The fungal colonization was investigated from14representative samples by using the method of dilution plating. The results of the survey showed that the number of yeast and mold were ranging from3.9×102to1.1×106cfu/g, the isolates were identified to belong to18species of5genera by morphology and means of sequencing the rDNA fragments, including8species of Aspergillus: Aspergillus niger、Aspergillus tubingensis、Aspergillus fumigatus、Aspergillus oryzae、Aspergillus sydowii、Aspergillus ochraceus、Aspergillus tamarii、 Sclerotium aspergillus;7species of Penicillium:Penicillium citrinum、 Penicillium chrysogenum、Penicillium oxalicum、Penicillium mallochii、 Penicillium chermesinum、Penicillium meleagrinum、Penicillium brocae;1species of Paecilomyces:Paecilomyces varioti;1species of Eurotium:Eurotium amstelodami and1species of yeasts:Stephanoascus ciferrii.Among of them, Sclerotium aspergillus、Penicillium chermesinum、Paecilomyces varioti、 Penicillium meleagrinum and Penicillium brocae were first isolated from dark tea.The dominant fungi of LiuPao tea was Ascomycota Eurotiomycetes Eurotiales Trichocomaceae, at the genus level, Aspergillus and Eurotium were predominant fungi, and the genus Penicillium and yeasts aeted as a subordinate partner according to the results of high-throughput sequencing. Combined with the classical culture-depended method, the genus Aspergillus、Penicillium、 yeasts and Eurotium were also dentified from the14tea samples, In all, this research provides theory support for further study on the isolates of LiuPao tea.
