Cephalosporin C, an important intermediate product of 7-aminocephalosporanic acid, is extracted from the fermentation broth, and the use of macroporous resin adsorption, desorption methods is directly related to the quality of 7-ACA, and also has crucial impact on cost.This paper examined the effect of preprocessing the original fermentation broth by ultrafiltration and vacuum drum methods, and both of the results were good. Titer of cephalosporin C ultrafiltrate and contents of various impurities, including the cephalosporin C free acid,were detected by HPLC method, and the change of p H value and the light transmittance of the fermentation broth and the supernatant were also studied.In this paper, the samples were collected, which were mainly produced by macroporous resin manufacturers at home and abroad. Under certain experimental conditions, two resins B3, B4 were selected. On this basis, impurities separation and adsorption saturation of the two resins were compared, and according to the scientifically credible data through parallel comparative experiments, we could draw conclusions: the properties of B3 resin were better than B4 resin on adsorption of resin, separation of impurities, desorption curve, cost and contamination resistance. Under current conditions, B3 resin was most suitable for the treatment of the cephalosporin C free acid stock solution. Depending on the selected resin, cephalosporin C ultrafiltrate as a research system, the processes of separation and purification of macroporous resin such as adsorption, washing and desorption were studied to determine the optimum feed p H value, optimum feed rate, adsorbent concentration and other operating conditions, which provided technical support to the production.
