Deacetyl Cephalosporin C (DCPC) is the main by-products in the production of Cephalosporin C (CPC). It is a structural analogue of the DCPC and accounts for about 20% of the total content of the CPC. Currently, after the separation and purification of the CPC from the fermentation broth, the fermentation broth of the CPC is directly released into the environment, which leads to pollution and Antibiotic residues in the environment. Research proved that the CPC can be converted to the pharmaceutical Intermediate,3- to acetyl -7-amino acid (D-7ACA) by chain hydrolysis, and the D-7ACA produced from DCPC has high yield and good quality characteristics in the producssion process of cefuroxime, cefoxitin and so on. Therefore, recovery of DCPC from CPC unbounded effluentwill contribute to both socity and economy.In this study, a new method of reverse phase high performance liquid chromatography for the quantitative analysis of DCPC was established firstly.Based on the results, the resolution DCPC and CPC was over 9, and the resolution of the CPC and DCPCc was over 3, the peak purity is more than 98%.The liner equation was Y=23176000X+ 178263.16, R2=0.9995, which showed good linear relationship from 0.05 to 0.9g.L-1. This method realizes baseline separation about DCPC and phenothiazine, CPC and DCPCc, providing reliable basis for subsequent quantitative analysis.Secondary, the stability of DCPC was investigated. It was found that the degradation of DCPC in low pH was very fast, especially when the pH less than 3, the degradation was more quickly. The temperature also had a great effect on the degradation of DCPC, the higher the temperature, the faster the degration of DCPC.So the separation and purification process of deathacetyl cephalosporin C should keep the low temperature.On the basis of above results a new process for the recovery of the DCPC from the CPC unbounded effluent was developed, the new process included steps of alkali precipitation, adsorption resin and ion exchange resion chramatrographie. The resin was screened by static adsorption experiments and the adsorption resin Aand ion exchange resin B were selected for the separation and purification of DCPC, and the two kinds of resins were all adsorption model. The sample pH of adsorption resin was 2.7, and the ion exchange resin was 4, ion exchange resin is mainly to get rid of DCPCc. The recovery rate of rangwthe DCPC from the two step resin can reach to about 74%, and the purity of the product can reach more than 90%. The purified solution was further purified by enzymatic hydrolysis, getting D-7ACA crystal powder with a purity of 98% and an absolute content of 95.6%.In this study,a new process for recovery of DCPC from CPC unbounded effluent was developed,the protein impurity was removed by alkali precipitation,and small molecular impurities was removed by adsorption resin and ion exchange resin chromatography,and relative high purity of DCPC was obtained,this word lay solid foundation on reducing the influence of antibiotics residual from CPC production on environment,and slso benefit for obtaining high-value Pharmaceutical Intermediate.
