| Fenugreek seed comprises seed coat endosperm and embryo. Seed coat contains essence, which can be used in cosmetic industry. Endosperm can be used as oil fracturing agent. Trigonelline can be used for against the cancer, diosgenin can be used as Hormone drugs,4-hydroxyl isoleucine for falling blood sugar. At present, most researchers extract the small molecules parts by crushing directly. Obviously, this can not make the comprehnsive utilization of fenugreek seeds. The thesis has carried out a series of discussion and research aimed at this situation, the main results are as follows:1. An analytical method using HPLC was developed for the trigonelline,4-hydroxyl isoleucine and diosgenin in Cotyledons, Under the optimize chromatographic conditions, the recoveries rate of the three kinds of compounds were about 98.96%,98.49%,98.17%. This method is simple, feasible and reproducible advantages.2. In this part, a study on experiment the effects of pressure, temperature and extraction time on oil yields extracted by SCF-CO2 was carried out, aiming to find out the optimal operation conditions. Taking GC-MS as the analytic method to measure the composition and content of the fatty acids in the Fenugreek seed oil. The measurement results were analyzed and discussed after the experiments in this part.3. Taking Fenugreek seeds as contrast experiment, and determine the distribution of small molecule compounds during the process mading of cotyledon. The results show that the trigonelline and 4-hydroxyl isoleucine are mainly distributed in boiling inorganic salt liquid and Cotyledons, Diosgenin are mainly distributed in Cotyledons. This date provide theoretical basis for amplification experiments.Extracting the small molecular compounds in cotyledon by 60% ethanol. And purificating the filtrate by ceramic membrane and organic membrane. A study on experiment the effects of pressure, temperature was carried out, aiming to select the key technological factors, provide data and research material for mass production. |
