Active Ingredient In Milk Thistle Extract

In this paper, effective composition was extracted from the silybum marianum Gaertn seed. Based on conventional organic solvent method, the extraction processes were done under ultrasonic or microwave in order to get high effect, save time, save energy and get new extraction methods for silybum marianum Gaertn total flavones and silybin. We studied on the extraction of effective composition from the silybum marianum Gaertn seed by conventional organic solvent method: the methods of cold exude, heat reflux and successive reflux with recover repeadly, the result shows that the method of successive reflux with recover repeadly is the best. Compared the several solvents: ethanol, methanol, ethyl acetate, acetone, acetonitrile, ethanol used as solvent can obtain the best silybum marianum Gaertn total flavones extracting effect and ethyl acetate is the best solvent for silybin. The experiments aimed at the optimum extractive parameters of silybum marianum Gaertn total flavones and silybin under ultrasonic or microwave pretreatment. Almost every factors'effecting on the extraction was analyzed and the ranges of them was determined, then the optimum extractive parameters under ultrasonic condition were obtained as follows: 60℃,60min, 100% ultrasonic power, 1:15 ratio of solid to liquid(m/v),the extraction yield of silybum marianum Gaertn total flavones was 60.09mg/g,and silybin was 37.26 mg/g;The optimum extractive parameters under microwave pretreatment condition were obtained as follows(20g defatted seed):汽化剂11mL, microwave pretreatment time 80s,extraction solvent 300mL,extraction time 40min,extraction tempreture 60℃,the　yield of silybum marianum Gaertn total flavones was 51.23 mg/g and shilibin was 23.97 mg/g. The method under microwave pretreatment was compared with the conventional method. Silybin was purified by recrystallization. Among three kinds of solvent (methanol, ethyl acetate-petroleum ether, acetone-petroleum ether) used for recrystallization, recrystallizing with methanol can obtain the best effect. In the end, the purified silybin was identified by color reaction, thin-layer chromatography, UV, IR. The result proved that the purified silybin was the pure silybin which we need.
