| The traditional Chinese medicine Radix Polygalae(Yuanzhi) has the efficacy of tranquilization, glaangal, eliminating sputum and detumescence. It is proved that it has more efficacy such as anticonvulsion, decompress, anti-mutation, against cancer and antibiotic, etc. The main components of Radix Polygalae are triterpenoid saponin, xanthone and alkaloid. Polygala Triterpenoid Acid(PTA) is the hydrolysis product of radix polygalae triterpenoid saponin. In order to researching the activity of radix polygalae triterpenoid saponin and development value, the thesis studied the preparation process and quality standard.Using triterpenoid saponin content as a marker, the optimum extraction technology was selected with the water experiment. The optimum extraction process is as follows: adding 8 times of water, dipping 20min, decocting 3 times, 120min for each time.The adsorption capability of macroporous resin on triterpenoid saponin was studied. The result indicates that the optimum purification process is as follows: D101 macroporous adsorption resin, medicine liquid concentration 0.05g/ml, flow rate 1 BV/h. Using elution ratio as the marker, the optimum elution method is as follows: eluted with distilled water, 3BV 30% ethanol and 3BV 70% ethanol in turn, flow rate 1BV/h. The purified triterpenoid saponin was collected in 70% ethanol eluting solvent.Using triterpenoid saponin content as a marker, the optimum concentrating and spray drying process is as follows: medicine liquid relative density 1.15 and temperature 50 ℃ , entrance temperature 140 ℃ , exit temperature 90℃. The obtainment ratio is 6.88% proved by three pilotscale experiments.Using polygalic acid content as a marker, the optimum hydrolysis process is as follows: 15 times of 10% hydrochloric acid, boiling 120min. The obtainment ratio is 3.11% proved by three pilotscale experiments.Using oleanolic acid as the standard substance, colorimetry method for the determination of content of total triterpenic acid in PTA was established.
