The nature medicinces is a very important portion of medicines. In china, thenature medicinces was also called Traditional Chinese medicinces (TCM), which arevery important for Chinese to prevent and cure disease all along. The components ofthe TCM are very complex, to study and apply the active ingredients of them, weneed to extract, separate and isolate the single component from plants. Therefore, theextraction, isolation and identification of the active components in TCM are more andmore important for the modernization of TCM.In this paper, a novel method to extract, separate and isolate the single componentfrom the natural products with the High-pressure homogenization extraction (HPHE)and high-speed counter-current chromatography was studied. The solanesol in potatoleaves and gentiopicrin, swertiamarin in Gentiana scabra Bunge was extracted andisolated. Operating conditions of extraction by HPHE and isolation by HSCCC wasoptimized.The mechanism of HPHE was discussed. There extraction method such as: routinesolvent extraction, ultrasonic extraction, HPHE was applied to extract solanesol inpotato leaves and gentiopicrin, swertiamarin in Gentiana scabra Bunge. The yield oftarget component was measured by HPLC. Compared with the routine extractions,efficiency using a novel HPHE was found to be improved in the results. In order tooptimize the HPHE operating conditions, including: pressure of HPHE, duration ofextraction and the material-solvent mixing ratio, a 3-factor-3-level (L9(33)) orthogonaltest was designed in this work. The best operating conditions of extraction of eachmaterial by HPHE was found.The HSCCC was applied to isolate solanesol in potato leaves and gentiopicrin,swertiamarin in Gentiana scabra Bunge, and the Operating conditions of HSCCCwas optimized. Using n-hexane-methanol (10:7, V/V) as two-phase solvent system,preparative HSCCC was successfully performed yielding 5 mg solanesol at 98.7%purity from 60 mg of crude extracts of potato leaves in a two-step separation. Fiftymilligrams of crude extracts of Gentiana scabra Bunge were separated using n-hexene-1-butanol- methanol-0.4% acetic acid in water (1.4:8:3:15.5, v/v) as thetwo-phase solvent system yielding 8 mg of swertiamarin, 18 mg of gentionpicrin, 11mg of trifloroside and 4 mg of an unknown compound with the purity of 96.7%,98.4%, 97.1% and 96.2%, respectively, in one-step separation.HPHE and HSCCC was a simple, rapid, and efficient method for extraction andisolation of target component from natural products. In this study we prove thefeasibility of using the HPHE and HSCCC to extract and isolate the target compoundfrom natural products. The method will be useful for extraction and isolation ofvarious other compounds from the natural products.
