Panax Ginseng. C.A.Mey is a kind of precious TCMs, its main active ingredient in ginseng is ginsenosides. Modern research shows that ginsenosides has anti-inflammatory, detoxifying, anti-fatigue, anti-tumor, anti-oxidation and vasorelaxant effects, Different ginenosides monomer has a lager differences in pharmacological function, so the research tendency about it is to use the ginsenosides monomer to cure related diseases. Therefore, research and development of ginsenosides has very important significance.At present, the mainly way to extract the TCMs still use the traditional extraction technology, such as impregnation, cooking method and so on, however, these methods generally exist disadvantages of the low extraction yield, long extration time and high energy consumption. To solve these problems, three new extraction techniques as microwave extraction> ultrasonic extraction and supercritical CO2fluid extraction were used to separate total ginsenosides in this paper,, then using spectrophotometric determinate its content, the results showed that the microwave method has the fast, safe and good extractive effect advantages. And get the best extraction conditions with70%ethanol as the solvent, extracted four times, each time5min, the solid-liquid ratio was1:12.High-speed counter-current chromatography(HSCCC) was used to prepare and separate ginsenosides from ginseng in this paper, under the conditions that the two-phase solvent system was composed of Ethyl acetate-n-butanol-water (1%acetic acid)-acetonitrile (7:3:10:1, v/v), lower phase was mobile phase, the speed was800r/min, separation temperature was25℃, flow rate was lml/min and the detection wavelength of254nm,29.3mg of Ginsenosides Rgl,13.4mg of Re,20.5mg of Rf from240mg crude extract of ginseng were obtained in one-step, the purity of these Ginsenosides was96.1%,98.4%and97.8%analyzed by HPLC and identified by ESI-MS.
